Abstract

The invention discloses an in-vitro culture and rapid propagation method of lithops. The method comprises the following steps: taking lithops leaves as explants, disinfecting the explants, then inoculating the explants to an initial culture medium and culturing for 3-4 weeks; inoculating the cultured and expanded explants into an induction medium and culturing for 6-7 weeks; selecting good calluses after induction culture, carrying out subculture multiplication culture, and culturing for 4 weeks; selecting strong and healthy multiple shoots after subculture multiplication culture, inoculatingthe multiple shoots into a rooting medium, culturing for 5 weeks, opening culture bottles and hardening the seedlings after the rooting rate reaches a certain number and the root length reaches the transplanting standard, then transplanting and managing tissue culture seedlings, and completing tissue culture and rapid propagation of lithops. The method is capable of rapidly and efficiently propagating lithops tissue culture seedling plants, increasing the survival rate and the rooting rate of the explants, producing a large quantity of high-quality regeneration seedlings in a short time, and improving the defects of slow growth speed and low propagation efficiency of the lithops.
