Problems solved by technology

[0003] Currently, the tissue-engineered nerve grafts used to repair nerve injuries are still mainly catheters with hollow lumens, which cannot accurately imitate the three-dimensional structure of normal nerves and the proportion of each component.
Decellularized neural scaffolds retain the three-dimensional structure of natural nerves while removing components such as cells, myelin sheaths, and axons, showing good clinical application prospects. However, the source of allogeneic decellularized nerves is still relatively limited.

Embodiment Construction

[0032] In order to make the above-mentioned features and advantages of the present invention more comprehensible, the following specific embodiments are described in detail in conjunction with the accompanying drawings.

[0033] This embodiment provides a specific preparation method of decellularized nerve grafts, specifically as follows:

[0034] 1. Material source: porcine sciatic nerve.

[0035] 2. Decellularized nerve graft preparation process:

[0036] 1) Obtain the sciatic nerve of an adult York pig from a mammal under sterile conditions, peel off and remove the fat and connective tissue around the nerve under a microscope.

[0037] 2) Peel off and remove the surrounding fat and connective tissue nerves under aseptic conditions, and take pictures and record them on a sterile sheet, such as figure 2 shown; the obtained nerve was cut into pieces and dissolved in ethanol, and its gas chromatogram was obtained in a gas chromatograph mass spectrometer, as shown in figure ...

Abstract

The invention discloses an acellular nerve graft which is an acellular heterogeneous nerve scaffold and contains a well-preserved three-dimensional orientation structure, which can guide nerve regeneration, of a nerve fiber nerve basilar membrane vessel. The structure has the components of cells, myelin sheaths, axons and fat that cause immunoreaction removed. The invention also discloses a preparation method of the acellular nerve graft, wherein the preparation method includes the steps: obtaining mammalian peripheral nerves under an aseptic condition, and stripping and removing fat and connective tissues around nerves under the aseptic condition; freezing and thawing the treated nerves repeatedly, carrying out decellularization by a chemical extraction and ultrasound combined method, cleaning with normal saline and adding deoxyribonuclease and ribonuclease solutions, and thus obtaining an initial acellular nerve graft; and freezing and drying the initial acellular nerve graft, then carrying out fat extraction treatment with supercritical carbon dioxide, and carrying out sterilization treatment to obtain the acellular nerve graft.

Description

technical field [0001] The invention designs medical biomaterials, and in particular relates to a decellularized nerve graft prepared by combining chemical and physical methods and a preparation method thereof. Background technique [0002] Peripheral nerve injury and nerve defect caused by injury are common diseases in clinical work, which not only have a serious impact on the life of patients, but also bring a huge social and economic burden. For peripheral nerve injuries with defects, autologous nerve transplantation is still the gold standard of treatment when tension-reducing sutures cannot be performed. However, the sources of autologous nerves are limited and cause secondary damage to patients. Therefore, for longer-distance nerve injuries, the application of tissue-engineered nerves and decellularized nerve grafts is a trend. [0003] Currently, the tissue-engineered nerve grafts used to repair nerve injuries are still mainly catheters with hollow lumens, which cann...
