Embodiment 1

[0026] Example 1 Wedelolide inhibits fungal-stimulated human macrophages from secreting IL-1β

[0027] 1. Experimental materials

[0028] 1.1 Experimental drug: wedelolide (purchased from selleck company)

[0029] 1.2 Experimental cells: human THP-1 cells (China Center for Type Culture Collection)

[0030] 1.3 Experimental fungus: 3.0772 strains of Aspergillus fumigatus (China Ordinary Microorganism Culture Collection Management Center)

[0031] 2. Experimental method

[0032] 2.1 Fungal preparation

[0033] The Aspergillus fumigatus strain was inoculated on the Sabouraud medium by the scratch method, and cultivated in a constant temperature incubator at 28°C for 2-3 days. Take 5ml of sterile PBS and put it into a petri dish, scrape the mycelium and conidia with a microbial scraping ring, collect the mixed solution, and filter the mycelium with sterile gauze. Centrifuge at 4°C and 4500rpm for 10 minutes, discard the supernatant, wash, and add 5ml of sterile PBS to the pel...

Embodiment 2

[0039] Example 2 The effect of wedelide on the corneal clinical score, pathological changes, Caspase-1 signaling pathway, and secretion of interleukin 1β (IL-1β) in fungal keratitis model mice, to explore the effect of wedelide combined with natamycin on the Effects of treatment in a mouse model of fungal keratitis.

[0040] 1. Experimental materials

[0041] 1.1 Experimental drugs: wedelolide (purchased from selleck company), natamycin eye drops (purchased from Alcon company)

[0042] 1.2 Experimental animals: C57BL / 6 mice (Jiangsu Changzhou Cavens Experimental Animal Co., Ltd.)

[0043] 1.3 Experimental fungus: 3.0772 strains of Aspergillus fumigatus (China Ordinary Microorganism Culture Collection Management Center)

[0044] 2. Experimental method

[0045] 2.1 Model mice

[0046] 8-week-old healthy and clean female C57BL / 6 mice were in good general condition. Slit-lamp microscope examination was performed before the experiment to rule out eye diseases, and one eye was r...

Abstract

The invention belongs to the technical field of pharmacy, and discloses application of wedelolactone in preparing a drug for treating keratomycosis. The wedelolactone has a therapeutic effect on the keratomycosis in vivo and in vitro conditions. Under the in vitro conditions, the effect of the wedelolactone on IL-1beta secretion is studied by a human macrophage model stimulated by aspergillus fumigatus. Under the in vivo conditions, a C57BL / 6 mouse keratomycosis model is adopted to study the effect of wedelolactone on corneal clinical scores, pathological changes, Caspase-1 signaling pathway and interleukin-1beta secretion in model mice. Furthermore, the application of the wedelolactone in combination with natamycin in preparing the drug for treating the wedelolactone is further provided.

Description

technical field [0001] The invention belongs to the field of pharmacy, and in particular relates to the application of wedelolide in the preparation of medicines for treating fungal keratitis. Background technique [0002] Fungal keratitis (FK) is an infectious corneal disease caused by fungi with a high rate of blindness. The patients are mainly farmers in developing countries. In recent years, due to the excessive use of hormones and antibiotics, long-term wear of contact lenses and the increase in the chance of corneal trauma, the incidence of fungal keratitis in some areas of my country has risen to the top of infectious corneal diseases, and most of the patients are working in the front line Young and middle-aged labor force. The treatment principle of fungal keratitis is early drug treatment, and those who cannot control the disease with drugs will eventually need surgery. Since the subjective symptoms of fungal keratitis are not obvious in the early stage, severe cor...
