Problems solved by technology

Since the tumor patients without mature sperm before puberty cannot realize the cryopreservation of their own sperm, and the cryopreservation of testicular tissue and spermatogonial stem cells is still in the stage of animal experiments, the fertility protection methods for this group are almost blank at present

Embodiment 1

[0023] 1. Establishment of Busulfan Azoospermia Model and Design of Animal Experiments

[0024] Inject busulfan intraperitoneally into C57 / BL6 male mice with a body weight of about 25 g (injection at 30 mg / kg body weight); busulfan is diluted according to busulfan in advance: DMSO: water = 20 mg: 5ml: 5ml, ready to use . After busulfan treatment, Substance P was injected into the tail vein once a week (at 5 nml / kg body weight) for a total of 10 weeks, and the changes in spermatogenic function were further detected.

[0025] 2. Isolation and culture of seminiferous tubules

[0026] Three-month-old C57BL / 6 male mice were sacrificed by cervical dislocation, and the testes were separated under sterile conditions. After rinsing with PBS containing 1% double antibody, remove the buffy film, add PBS, blow off the small tube with a pipette, the small tube naturally settles in PBS, let stand to remove the supernatant, repeat 4 to 5 times, and pick the small tube to the culture plate ...

Abstract

The invention discloses application of a P substance in preparation of a drug for treating male infertility. Treatment of an azoospermia mouse model by the P substance finds that the P substance can effectively improve spermatogenic functions of the azoospermia mouse model caused by busulfan and further proves that the P substance improves the spermatogenic functions by promoting proliferation ofspermatogenic cells. The invention provides a new thought and theoretical support for treatment of male infertility.

Description

technical field [0001] The invention relates to the use of SP (Substance P, Substance P) in the preparation of medicines for treating male infertility. Background technique [0002] With the prominence of the global decline in fertility, "male fertility protection" has become a hot topic of research by experts in related fields around the world. Decreased sperm quality is an important reason for male fertility decline. According to WHO statistics, the decline in sperm quality has affected 7% of males of reproductive age in the world. In the past 40 years, the overall sperm concentration has dropped by 59.3% (see: Schlegel, P.N. (2009) Evaluation of male infertility.
