The method comprises the steps: submerging roots of the rhubarb young seedlings into sterile digested effluent, carrying out activating treatment for 7 to 14 days under the conditions that a temperature is controlled to 20 DEG C to 24 DEG C, the intensity of illumination is controlled to 2,200lx to 2,800lx, and the duration of solar radiation is 14 to 15 hours, then, taking out the rhubarb young seedlings, placing the rhubarb young seedlings at a shady and cool ventilating place, covering the rhubarb young seedlings with clean cotton fabrics, sprinkling sterile water at any time, and carrying out seedling airing for 2 to 3 hours; inoculating a seedling rejuvenating culture medium with the young seedlings at an inoculum density of 80 plants per square meter to 100 plants per square meter under the standard that the roots of the young seedlings are completely submerged into the culture medium, then, sealing the culture medium, controlling a day temperature to 22 DEG C to 24 DEG C and a night temperature to 14 DEG C to 15 DEG C, and controlling relative humidity of air to 50% to 70%, duration of solarradiation to 15 to 17 hours and intensity of illumination to 2,200lx to 2,800lx; and spraying an aqueous solution of salicylic acid, polysaccharides and FeSO4 or an aqueous solution of jasmonic acid,the polysaccharides, the FeSO4 and calcium superphosphate to unsealed stems and leaves at intervals of 2 days, cleaning leaf surfaces with sterile water before spraying at a time, and carrying out seedling rejuvenating culture for 12 to 15 days.
