Embodiment 1

[0048] The black soil used in this example was collected from the Hailun Ecological Experimental Station of the Chinese Academy of Sciences, and the albic soil was collected from the Sanjiang Plain. Both soils were cultured at 25°C for more than 24 hours, and the soil water content was 20% (air-dried soil weight). The catalase activity of the two soils was detected by the above method.

[0049] The specific analysis steps for each soil are as follows:

[0050] 1) Weigh 4 parts of 5g fresh soil into a 100ml Erlenmeyer flask (n≥2), add 5ml 10% sulfuric acid and 5ml 10% trichloroacetic acid to the control soil to inactivate catalase; Put the Erlenmeyer flask with unpassivated soil in a 0℃ ice bath. After 15 minutes, add 10ml of 1% hydrogen peroxide solution to each Erlenmeyer flask, carefully mix the reactants, and incubate at the same temperature for 1 hour;

[0051] 2) After the cultivation, add 5ml 10% sulfuric acid and 5ml 10% trichloroacetic acid to the test bottle to inactivate...

Embodiment 2

[0063] The brown soil used in this example was collected from the Shenyang Ecological Experimental Station of the Chinese Academy of Sciences, and the cinnamon soil was collected from Chaoyang. The water content of the two kinds of soil is 20% (air-dried soil weight), cultured at 25°C for more than 24 hours, and the catalase activity is determined. The specific steps are as follows: Substrate H 2 O 2 The concentration is 2%, 5 mL is added, and the remaining steps are the same as in Example 1.

[0064] The experimental results are as follows:

Abstract

The invention relates to an analytical method for measuring catalase activity in soil, which comprises the following steps 1) weighing out n parts of fresh soil and placing the parts in 100 ml triangular flasks, passivating the catalse by adding sulphuric acid and trichloroacetic acid to control soil, keeping all soil samples in 0 DEG C ice bath for 15 minutes, adding catalase solution to the triangular flasks; mixing and culturing the mixture at the same temperature for 1 hour; 2) passivating the catalse by adding sulphuric acid and trichloroacetic acid to the flasks; 3) shaking all triangular flasks and filtering; 4) preparing a tube containing a titanic sulfate solution for each sample, adding a certain amount of filtrate and distrilled water and mixing until the fluid becomes yellow; 5) detecting absorbance by comparing color on a spectrophotometer; 6) calculating the amount of hydrolyzed catalase. The method has the advantages of 1) simplified steps and reduced uncertainty in the experiment; 2) high accuracy and easy operation; 3) reliable result and good reproducibility.

Description

Technical field [0001] The invention relates to the determination of catalase activity in soil, in particular to an analysis method for detecting catalase activity in soil. Background technique [0002] Catalase in the soil can enzymatically decompose hydrogen peroxide into water and molecular oxygen. The specific reaction is as follows: [0003] [0004] Hydrogen peroxide is formed in the process of biological respiration and as a result of various biochemical reactions of organic substances. In organisms, and possibly in soil, catalase acts to destroy hydrogen peroxide, which is toxic to organisms. Therefore, catalase in the soil indirectly affects the level of soil fertility, and its activity is strongly affected by soil conditions such as moisture, temperature, and soil texture. It is also affected by farmland farming systems, management measures, and different crops. Therefore, the test of catalase activity in soil is of great significance. So far, the determination of cat...
