Problems solved by technology

[0004] Most of the traditional detection methods for hepatitis C virus antibody use the enzyme-linked immunosorbent method with materials such as polystyrene as the carrier. This method is relatively simple, but the disadvantage is that the detection sensitivity is poor, and it is prone to missed detection.

Embodiment Construction

[0030] The method for detecting hepatitis C virus antibody using magnetic particles as a carrier of the present invention, the main steps include:

[0031] (1) Fixing: using magnetic particles as the carrier for reaction and separation, the hepatitis C virus antigen is coupled on the surface of the magnetic particles by the following method:

[0032] Take 30ul of magnetic particles, wash with 0.02MPBS, PH7.6 buffer (i.e. phosphate buffer) for 1 to 3 times;

[0033] Dissolve 25% v / v glutaraldehyde into the above buffer solution containing magnetic particles at a ratio of 1:10, mix well and place in a shaker, shake at 20-40°C, 180r / min, fully react for 2 hours, magnetic Separating, discarding the supernatant, and washing the magnetic particles twice with a buffer;

[0034] Dissolve the hepatitis C virus antigen in the buffer solution containing the above-mentioned processed magnetic particles at a ratio of 1:2K, mix evenly, place in a shaker, shake at a medium speed under the c...

Abstract

The present invention discloses a method for detecting third type hepatitis virus antibody by using magnetic micro-particle as transporting species which includes steps as follows: (1) using magnetic micro-particle as transporting species for reacting and separating, coupling third type hepatitis virus antibody on the magnetic micro-particle surface; (2) adding into confining liquid, oscillating 10 min with middle speed under condition of 20-40 deg c, magnetic separating, giving up supernatant; (3) combining the third type hepatitis virus antibody packed on the magnetic micro-particle surface with the specificity antibody on waited blood; (4) separating the magnetic micro-particle with specificity antigen-antibody complex on surface through an externally-applied magnetic field; (5) combining the antibody complex on the magnetic micro-particle surface with enzyme label rat mouse-anti-body IgG; (6) using chemiluminescence detector, irradiancy substrates A liquor and B liquor for detecting sample irradiancy value. The method has characteristics of rapid detecting speed, high specificity, stabilising, no radioactive pollution which can increase sensitivity and stabilizing ability for detecting.

Description

technical field [0001] The invention relates to the field of immunological detection, in particular to a method for detecting hepatitis C virus antibody using magnetic particles as carriers. Background technique [0002] Hepatitis C (HCV) is a global epidemic and is the main cause of end-stage liver disease in Europe, America, Japan and other countries. According to the statistics of the World Health Organization, the global HCV infection rate is about 3%. The source of infection of hepatitis C is mainly acute clinical type and asymptomatic subclinical patients, chronic patients and virus carriers. HCV is mainly transmitted through blood, and the main routes are blood transfusion, blood products, and damaged skin and mucous membranes. my country's serum epidemiological survey data show that the general population anti-HCV positive rate is 3.2%. The anti-HCV positive rate increased gradually with age, from 2.0% in the 1-year-old group to 3.9% in the 50-59-year-old group. ...
