Embodiment 1

[0045] Method flow see figure 1 .

[0046] 1. Collection of calibration set samples:

[0047] Collect 22 batches of 20 parts of the first extraction of Salvia miltiorrhiza, 17 parts of the second extraction, 14 parts of the third extraction, a total of 51 samples of the salvia miltiorrhiza extract, as a calibration set sample;

[0048] 2. Determination of the reference value of the calibration set sample:

[0049] Prepare the test solution and reference solution according to the phosphomolybdotungstic acid / casein method in Appendix XB of the Chinese Pharmacopoeia in 2005, determine the tannin content according to the law, and uniformly control the color reaction time to 30min. The distribution range of the measured tannin content in the sample set was 11.2-197.1 μg / ml.

[0050] 3. Calibration set sample spectral data collection:

[0051] Near-infrared spectra of calibration set samples were collected using ANTARIS Fourier transform near-infrared spectrometer (Thermo Nicole...

Embodiment 2

[0071] According to the method of Example 1, the difference is that a principal component regression (PCR) method is used to establish a tannin quantitative correction model.

[0072] 1. Select the same calibration set sample data as in Example 1.

[0073] 2. Establishment of calibration model

[0074] Before establishing the calibration model, first use the computer custom software (Unscrambler9.6) to optimize the preprocessing method and band selection of the spectrum, and process the original data with the optimal band range and sample preprocessing method to obtain the characteristic spectral information of the Danshen extract. The quantitative calibration model of tannin was established by principal component regression method, and the internal validation was carried out by leave-one-out full crossover method.

[0075] (1) Modeling and optimization of near-infrared spectral data in different bands:

[0076] Take the full wavelength of the sample set, 4327-4898cm -1 、55...

Abstract

The invention provides a method for measuring the content of tannin in a red sage root extracting solution by a near infrared spectrum, which comprises the following steps: firstly, collecting typical red sage root extracting solutions to be used as a correcting sample set; scanning to acquire a near infrared spectrum chart of the correcting sample set; selecting a proper spectrum waveband and a proper pretreatment method to acquire the characteristic spectrum information of the red sage root extracting solution; by using the result of the content of the tannin in the sample set measured by a phosphomolybdium tungstic acid / casein method as a reference value, constructing a multivariate correcting model of relation between the near infrared characteristic spectrum information and the tannin content of the red sage root extracting solution by applying the chemometrics technology; carrying out near infrared spectrum scanning on the red sage root extracting solution sample with unknown content, and acquiring the characteristic spectrum information; and rapidly calculating the content of the tannin by utilizing the constructed quantitative correcting model. The invention has the characteristics of simple method, rapidness and high accuracy, can effectively solve the problem of monitoring of the content of the tannin in the red sage root extracting solution to ensure the quality of red sage root injections, and can be popularized and applied in production of traditional Chinese medicine preparations.

Description

technical field [0001] The invention belongs to a method for monitoring the production quality of traditional Chinese medicines, in particular to a method for quickly measuring the tannin content in Danshen extract by near-infrared spectroscopy. Background technique [0002] Tannins are a class of water-soluble polyphenolic compounds widely present in Chinese herbal medicines, which have the function of precipitating proteins. Tannins in traditional Chinese medicine preparations will not only affect the stability and clarity of the preparations, but may also cause a series of serious physiological reactions. For example, some tannins injected into the body will cause a series of clinical symptoms such as jaundice and liver necrosis. Therefore, tannins are often removed as impurities during the preparation of traditional Chinese medicines. In recent years, the safety of traditional Chinese medicine injections has attracted widespread attention. Tannins in traditional Chinese...
