Embodiment 1

[0048] 1. Calibration set samples: Use the method of GB / T 22966-2008 and GB / T 21312-2007 to determine the negative milk samples as the milk matrix of the calibration set. Dafloxacin at 7 concentration levels and sulfamethoxazole standard working solution at 8 concentration levels were added, the two concentrations of antibiotics were randomly mixed, and a total of 15 samples were used as a calibration set.

[0049] 2. Determination of calibration set samples: pipette 5mL of milk into a 50mL volumetric flask, add random combinations of 0, 10, 20, 30, 40, 50, 60 μg / L danofloxacin and 0, 30, 50, 100, 120, 150, 200 μg / L Sulfamethoxazole standard solution, add ultrapure water to the mark, mix well, take 8mL sample solution and place it in a 10mL centrifuge tube, add 0.5mL 30% (v / v) trichloroacetic acid (TCA) solution , vortex, let stand for 10 minutes, centrifuge at 4000r / min for 10 minutes, and filter. Take 5 mL of the filtrate in a test tube, add 400 μL of fluorescamine acetone ...

Abstract

The invention relates to a method for determining residual Sulfamethoxazole and Danofloxacin in milk simultaneously by a fluorescence spectrum. The method includes scanning deproteinized and derivatized milk according to a three-dimension excitation emission matrix fluorescence spectrum, analyzing the fluorescence spectrum by means of chemical metering, and establishing a multi-variate correction model capable of detecting content of Sulfamethoxazole and Danofloxacin in milk simultaneously quickly. By the fluorescence method, negative samples and positive samples can be identified quickly when in prediction of the residual Sulfamethoxazole and Danofloxacin in unknown milk samples by the established correction model, and further, content of two drug residues can be determined. The fluorescence method has the advantages of simplicity, rapidness and high flexibility, detection efficiency is increased greatly, operation processes for detecting antibiotic residues are simplified, samples in batch can be processed, and the fluorescence method can be popularized in detection application.

Description

technical field [0001] The invention relates to a detection method for simultaneous determination of two antibiotics in milk, in particular to a fluorescence method for simultaneous determination of sulfamethoxazole and danofloxacin in milk. Background technique [0002] In recent years, with the improvement of the income of Chinese residents and the change of consumption concept of dairy products, the dairy industry of our country has developed rapidly. However, judging from the current production and processing status of domestic dairy industry, the quality and safety status of dairy products in my country is not optimistic. The level of farm farming of dairy cows in my country is low, and most of the raw milk comes from dairy cow farmers, so the quality of raw milk fluctuates greatly. The adulteration of dairy products, the abuse of antibiotics and many other issues have caused major hidden dangers in the safety of dairy products. [0003] Sulfamethoxazole and dafloxaci...
