The method comprises the following steps of: drying matsutake, smashing, extracting with boiled water for 2-5 hours, centrifuging extracting a supernatant, filtering the supernatant, and concentrating to obtain a concentrated solution, wherein the mass ratio of the supernatant before concentration to the supernatant after concentration is (10-25):1; performing alcohol precipitation on the concentrated solution, centrifuging, extracting a precipitate, and performing freeze drying to obtain freeze-dried powder; and dissolving the freeze-dried powder into deionized water, adding ethanol of which the volume percentage is over 90 percent till the volume percentage of the ethanol is 60 percent, performing alcohol precipitation at the temperature of 4 DEG C for 6-10 hours, centrifuging and extracting a precipitate.
