Abstract

The invention relates to the technical field of biomedicine, in particular to a novel target resistant to Japanese encephalitis virus (JEV) infection and application. Human neuroblastoma cells (SK-N-SH) are taken as target cells, RNA technology is adopted to reduce expression of host protein of the target cells to find host factors capable of effectively inhibiting JEV-infected human neuroblastoma cells (SK-N-SH) so as to protect functions of a nerve system and prevent virus from infecting a central nerve system to cause inflammation. The invention provides application of Cofilin in preventing and treating JEV infection, and experiments find that Cofilin plays an important role in JEV-infected SK-N-SH, and JEV infection can be inhibited remarkably by reducing expression of Cofilin. The invention further provides application of Cofilin in preparing drug for preventing or treating JEV infection.

Description

technical field

[0001] The invention relates to the technical field of biomedicine, and is a new target and application for resisting Japanese encephalitis virus infection. Background technique

[0002] Japanese encephalitis virus, also known as Japanese encephalitis virus (JEV), belongs to the Flaviviridae (Flaviviridae) genus (Flavivirus), and is a single positive-strand RNA virus with an envelope [Gubler, D., G.Kuno , and L. Markoff. Flaviviruses, p. 2007. 1153-1252. In D. M. Knipe and P. M. Howley (ed.), Fields virology, 5thed., vol. 1. Lippincott-Williams & Wilkins, Philadelphia, PA.]. JEV is the pathogen of Japanese encephalitis, which is mainly transmitted by mosquitoes. After infection, it can cause severe acute neurological symptoms such as encephalitis or meningitis, which poses a huge threat to human health. Japanese encephalitis is mainly prevalent in parts of East Asia, Southeast Asia, and Oceania. Currently, the main means of control are vaccine prevention and...

Examples

Embodiment 1

[0045] 1 Design and synthesis of specific siRNA sequences for each host cell molecule.

[0046] 1.1 For each target gene, search NCBI GeneBank to obtain the full sequence and mRNA sequence, use existing network resources and commonly used software to conduct biological analysis of each target gene, and select the coding region as the target sequence for siRNA design. Refer to the siRNA design principle, and compare it with the human genome sequence through the blast function of the GeneBank database to ensure no homology; exclude potential siRNAs with 8 consecutive bases at the 5' end of the antisense chain paired with other genes; exclude any consecutive 14 bases base-pairing potential siRNAs with other genes. The design software was used for pre-evaluation and determination, and three targets with the best kinetic parameters were selected to enter the subsequent experimental process. A total of three interference sequences were synthesized for each gene, as shown in Table 1....
