Problems solved by technology

[0004] At present, there are few studies on molecular markers of resistance to WSSV in Litopenaeus vannamei, and there is a lack of available markers for the cultivation of molecularly assisted disease-resistant strains. Therefore, this study analyzed the SNP in an ALF gene (nLvALF6) of Litopenaeus vannamei Markers, and use the method of association analysis to screen out the molecular markers in the gene that are significantly related to resistance to WSSV, and provide effective markers and technical means for molecular marker-assisted breeding of disease-resistant populations

[0023] Acquisition of SNP markers related to shrimp resistance to WSSV

[0024] 1. Acquisition of WSSV susceptible and resistant materials

[0025] WSSV susceptible and resistant materials come from the mixed population of "Kehai No. 1" multi-lineage. 500 healthy Litopenaeus vannamei were selected and placed in 12 50L aquariums. Before the experiment, 10 prawns were randomly selected to detect no WSSV infection. The experimental water temperature was (23±1)℃. After the starvation treatment, the diseased shrimps caused by WSSV infection were fed (WSSV virus content was 10 4 -10 6 copy / ng DNA). After challenge, check 3 times a day (each interval 6h) and collect dead shrimp, randomly sample and extract genomic DNA, the earliest dead 48 prawns were used as WSSV susceptibility group (susceptible group, sus). On the 14th day after challenge, the survival of Litopenaeus vannamei was stable, and 48 surviving prawns were selected as the WSSV resistant group (res).

Abstract

The invention discloses an anti-WSSV (white spot syndrome virus) candidate molecular marker from an anti-lipopolysaccharide factor gene (nLvALF6) of litopenaeus vannamei and application of the anti-WSSV candidate molecular marker. An SNP marker significantly correlated to anti-WSSV in the anti-lipopolysaccharide factor gene (nLvALF6) of the litopenaeus vannamei is an SNP locus (nLvALF6g-258-A>T) at the position 258bp in SEQIDnLvALF6-g. An effective marker is provided for vannamei disease-resistant molecule breeding, an important guidance is provided for application of the disease-resistant molecular marker to genetic breeding, and the anti-WSSV candidate molecular marker has very good application prospects in the study of vannamei anti-WSSV variety breeding.

Description

technical field [0001] The invention relates to the technical field of aquatic animal molecular genetics and aquatic animal germplasm innovation, and relates to an anti-WSSV candidate molecular marker derived from Litopenaeus vannamei anti-lipopolysaccharide factor gene (nLvALF6) and its application. Background technique [0002] Anti-lipopolysaccharide factor (anti-lipopolysaccharide factor, ALF) is a small antibacterial peptide from crustaceans, which was first identified from the blood cells of Limulus polyphemus, and has obvious anti-rough Gram-negative bacteria and anti- endotoxin effect. The results of functional studies show that crustacean ALF has broad-spectrum and potent antibacterial effects (Beale K M, Towle D W, Jayasundara N, Smith CM, Shields JD, Small HJ, Greenwood SJ.
