Problems solved by technology

Due to the tall cotton plants, if the temperature-sensitive characteristics of this temperature gradient are studied, a large-scale temperature-controlled artificial climate chamber is required, and the construction and operation costs are huge.

Embodiment 1

[0040]Plant the cleistogamous pollination material CJ1548143 and the normal flowering control material Zhongmiansuo 63 in the experimental field. During the flowering period, at about 5:30 every morning, use scissors to cut the three fruit branches of the two materials that contain the flower buds that are about to open that day, and cut them off. The flower buds to be opened and the parts other than the opposite leaves are placed in conical flasks filled with sterile water and cultivated in a temperature-controlled light incubator. The light intensity of the light incubator was constant (17600 lux), the temperature range was 16-38°C, and the temperature gradient was 1-2°C. It started at 6:00 and ended at 18:00. The opening of the petals was observed and recorded every 2 hours.

[0041] The obtained results are shown in Table 1.

Embodiment 2

[0048] Plant the cleistogamous pollination material CJ1548143 and the normal flowering control material Zhongmiansuo 66 in the experimental field. During the flowering period, at about 5:30 every morning, use scissors to cut the fruit branches of the two materials that contain flower buds that are about to open that day, and cut off the fruit branches that are about to open. Flowers and the parts other than the opposite leaves were placed in Erlenmeyer flasks filled with sterile water and cultivated in a temperature-controlled light incubator. The light intensity of the light incubator is constant (17000lux), the temperature range is 24-38°C, the gradient between different temperatures is 1-2°C, and each temperature has 3 fruit branches, starting from 6:30 and ending at 18:30, Observe and record the opening of the petals every 2 hours.

[0049] The obtained results are shown in Table 2.

Embodiment 3

[0056] Plant the cleistogamous pollination material CJ1548143 and the normal flowering control material Zhongmiansuo 66 in the experimental field. At 6:00 every morning during the flowering period, use scissors to cut the fruit branches of the two materials that contain the flower buds that are about to open that day, and cut off the flowers that are about to open and the parts other than the paraphyll leaves were placed in Erlenmeyer flasks filled with sterile water and cultivated in a temperature-controlled light incubator. The light intensity of the light incubator is constant (18000lux), the temperature range is 24-38°C, the gradient between different temperatures is 1-2°C, and each temperature has 4 fruit branches, starting at 6:30 and ending at 17:30, Observe and record the opening of the petals every 1-2 hours.

[0057] The obtained results are shown in Table 3.

Abstract

The invention relates to an identification field of cotton thermosensitivity, and particularly relates to a method for identifying thermosensitivity of cotton clistogamy. The method includes steps of collecting fruit spur with buds to be bloomed in the same day of the clistogamy material, respectively performing light cultivation for over 4 hours under different temperatures; observing the bud blooming under different temperature conditions, and acquiring the stable flower closing temperature scale of clistogamy, blooming critical temperature of clistogamy material, and complete blooming temperature of clistogamy material. Through cultivating the collected fruit spur with buds to be bloomed in the same day under different temperatures for a certain time, the other cultivating conditions such as culture mediums and light intensity are the same; then the bud blooming conditions under different temperature conditions are observed, thus the temperature scale of the stable flower closing, blooming critical temperature and complete blooming temperature of the clistogamy material are acquired. The method is simple in operation, low in cost, and can rapidly identify the flower closing and blooming thermosensitivity of cotton clistogamy.

Description

technical field [0001] The invention relates to the field of identification of temperature-sensitive characteristics of cotton, in particular to a method for identifying the temperature-sensitive characteristics of cotton cleistinia pollination materials. Background technique [0002] Cotton is a frequent cross-pollinated crop, and the natural outcrossing rate is generally 3-20%, and the highest can reach 50%. At present, conventional species and hybrid species have been widely promoted in production. In the process of self-propagation of conventional species and self-propagation of hybrid species, due to the certain outcrossing rate of cotton, outcrossing is easy to occur, resulting in biological mixing and degrading the characteristics of excellent varieties. In view of this, breeders and seed companies need to invest a lot of manpower, material and financial resources every year to isolate the conventional cotton varieties and hybrid parents or artificially self-bred the...
