Abstract

The invention provides a construction method of RFF1 cells and belongs to the technical field of biology, aiming to construct a kind of RFF1 cells for cellular immunotherapy, which are super T cells with both attack and defense, with good precision and high killing rate. The construction method includes the steps of loading tumor mutated polypeptides on PBMC (peripheral blood mononuclear cell) andthen performing polypeptide shocks on the PBMC loaded with the polypeptides; performing expanding culture after shocks to obtain EF cells; taking the tumor mutated polypeptides as antigen to direct stimulate the EF cells to screen precise polypeptides; knocking out immunosuppressive signal molecules on the PBNC based on the CRISPR (clustered regularly interspaced short palindromic repeats) technique; loading the PBMC with the precise polypeptides which are mixed with the PBMC knocking out the immunosuppressive signal molecules for co-culture, and during the co-culture process, performing multiple shocks with the precise polypeptides to obtain the RFF1 cells. The construction method can be applied to the cellular immunotherapy technique.
