Problems solved by technology

In the current research, there is no precedent for constructing an ITP animal model with primates, especially cynomolgus monkeys, using ITP-specific antigens, especially CD61 protein, as an immunogen. Compared with other animal models, it is more effective It is similar to the incidence of human beings, and has more specificity of inducing the onset, which is helpful for in-depth research on the pathogenesis of ITP and further application in the preparation of drugs for the treatment of ITP

Embodiment 1

[0016] Example 1: Induction of ITP animal model.

[0017] Experimental animals: cynomolgus monkeys (Cynomolgus), male or female, aged 2.5-4.0 years, weighing 2.5-4.5kg.

[0018] Experimental environment: The test monkeys will be placed in stainless steel monkey cages (800 mm × 700 mm × 820 mm) in Pengli Biological Animal Room. The adaptation period is: 2 monkeys per cage; the experimental period and postoperative care are: 1 (with equipped animal welfare appliances). The room number in which the animals were housed throughout the experiment will be recorded in the experiment record. The room where the test monkeys are placed is located in the Pengli biological animal room, and the number of filtered ventilation in this area is 10-20 times per hour. Temperatures will remain between 20-26°C (68-79°F) and relative humidity 40-70%. Continuously observe and record temperature and humidity. The lighting conditions were 12 hours (08:00-20:00) daylight irradiation and 12 hours no ...

Embodiment 3

[0023] Example 3: Detection of ITP animal models.

[0024] Detection Indicator:

[0025] 1) Body weight: before the start of the experiment, measure and record the body weight of the animal;

[0026] 2) Blood collection: before immunization, from 7 days after immunization, venous blood collection twice a week, EDTA anticoagulant, about 200ul, up to 28 days;

[0027] 3) Complete blood count: the frequency is the same as that of blood collection, no significant decrease in platelets was observed within three weeks, and the animals were given a second antigen immunization on Day 7-28, and the method was the same as the first time;

[0028] 4) Serum sample preparation: the frequency is the same as that of blood collection, the serum is separated to about 400ul, and stored at -80°C;

[0029] 5) Determination of bleeding time: several time points before immunization and when a significant decrease in platelets is found.

Abstract

The invention belongs to the field of biology, and in particular relates to a method for constructing a monkey ITP model induced by a platelet-specific antigen. In particular, it provides a method for obtaining an ITP model by immunizing monkeys with CD61 protein as a specific antigen. Including the application of CD61 protein in the preparation of animal models of thrombocytopenic purpura, the sequence of the CD61 protein is shown in SEQ ID NO:2. The induction method of the invention can effectively induce thrombocytopenia and purpura symptoms, and obtain a stable monkey ITP model.

Description

technical field [0001] The invention belongs to the biological field, and relates to a method for making an animal model of idiopathic thrombocytopenic purpura (ITP), in particular to a method for making a monkey ITP model induced by a platelet-specific antigen. Background technique [0002] Immune thrombocytopenic purpura (ITP) is an autoimmune disease in which the decrease in peripheral blood platelets leads to bleeding in the skin, mucous membranes and internal organs. The etiology is mainly the increased destruction of platelets mediated by anti-platelet autoantibodies or cytotoxic T cells in patients. When the platelet count is extremely low (Plt<5×10 9 ) may cause fatal complications such as subarachnoid space, cerebral hemorrhage, gastrointestinal bleeding and other parts of the bleeding. For the clinical treatment of ITP patients, only a small number of patients can be completely relieved (Plt>100×10 9 and no bleeding), most patients experience a chronic rec...
