Disclosed are a method for determination of HDL cholesterol, a reagent for the determination and a kit for the determination which realize a convenient and accurate determination of HDL cholesterol. A method for determination of cholesterol in a high-density lipoprotein in a sample, the method comprising the steps of reacting the sample with i) a cholesterol ester hydrolase and a cholesterol oxidase or ii) a cholesterol ester hydrolase, an oxidized coenzyme and a cholesterol dehydrogenase in an aqueous medium containing a specific nitrogenated surfactant having a structure of an amine or ammonium salt and a polyanion to produce hydrogen peroxide or a reduced coenzyme, and determining hydrogen peroxide or the reduced coenzyme.
