Embodiment 1

[0013] 1. Test material

[0014] Blood samples from 118 first-generation Bian chicken hens were collected from the Institute of Animal Husbandry and Veterinary Medicine, Shanxi Academy of Agricultural Sciences. The side chicken groups taken come from the same batch, reared in single cages, with consistent management and nutritional levels. Heparin sodium was used as an anticoagulant, and 0.5 mL of blood samples were collected from wing veins. Genomic DNA was extracted by conventional phenol-chloroform method, and the concentration of DNA was determined for later use.

[0015] 2. Primer Design and PCR Amplification

[0016] 2.1 Design of enzyme digestion primers

[0017] According to the chicken MSTN gene sequence in GenBank (GenBank accession number is AF346599), a pair of primers were designed for the BbvI site of exon 1, and the size of the amplified product was 162 bp. The primer sequences are as follows:

[0018] P1: F: 5'-GCATTAGCAGGGACGTTAT-3'; (SEQ ID NO: 1)

Abstract

The invention relates to the field of poultry breeding, and in particular relates to a method for screening the number of eggs produced by Bian chicken through DNA (deoxyribonucleic acid) labeling. The method comprises the following steps: extracting the chicken genome DNA of a sample to be detected; amplifying with a specific primer to obtain a target segment of an MSTN gene; carrying out enzymedigestion on the target segment with restriction enzyme BbvI; after gel electrophoresis, carrying out silver staining development so as to obtain an enzyme digestion map of DNA; and judging the number of the eggs produced by the sample to be detected according to the quantity and size of strips in the map. The detection method selected by the invention is simple and rapid, is not influenced by external environment, and can be used for molecular genetic labeling of the number of the eggs produced by Bian chicken to carry out marker-assisted selection.

Description

technical field [0001] The invention relates to the field of bioengineering, in particular to a method for rapidly increasing the number of eggs laid by side chickens by using DNA markers. Background technique [0002] Traditional quantitative genetics’ understanding of the genetic mechanism of quantitative traits is based on the abstract assumption of micro-effect polygenes, and treats all genes that affect a quantitative trait as a whole, regardless of how each gene works . This hypothesis has played an important role in many years of animal and plant breeding and promoted the progress of animal and plant breeding. It was a very good foundation under the theoretical and technical conditions at that time. However, this hypothesis does not understand the genetic background of quantitative traits, the location of genes controlling quantitative traits on chromosomes and their transmission rules, and cannot make accurate estimates of their effects, let alone isolate and locate...
