Problems solved by technology

Whether anesthesia itself has a protective effect on rats with cerebral infarction is controversial. Therefore, anesthesia assists in cooling, which interferes with the results of animal experiments and affects the neurobehavioral evaluation.

Embodiment 1

[0027] 1. Experimental method

[0028] The 35 test rats were divided into a mild hypothermia group and a normal temperature group, and the former was divided into a 2h mild hypothermia group and a 6h mild hypothermia group according to the different time points of mild hypothermia administration. All rats underwent surgery before hypothermia or normothermia intervention (needed for animal experiments).

[0029] 1. Sub-low temperature implementation

[0030] Rats to be subjected to mild hypothermia were placed in a ventilated environment at 4°C, and ice packs were used to assist in cooling. Self-made ventilated mouse cages can avoid direct contact between rat skin and ice packs and avoid frostbite. At the specified time point 2h or 6h, put the rats in the mild hypothermia group into the mouse cage, place them in a low temperature environment of 4°C, put ice packs outside the mouse cages to assist in cooling, the target temperature is 33±0.5°C, and continuously observe the cha...

Abstract

The invention relates to a preparation method for a rat mild hypothermia animal model. The preparation method comprises the steps as follows: placing a rat into a ventilated rat cage, placing the rat cage into a low temperature environment of 4 DEG C, placing an ice bag outside the rat cage for auxiliary cooling, and continuously monitoring the change of the anal temperature of the rat based on the target temperature of 33 plus or minus 0.5 DEG C; and when the anal temperature is lower than the lowest limit of 32.5 DEG C, moving away the ice bag or placing the rat and the rat cage into a room temperature environment, and when the anal temperature is higher than the highest limit of 33.5 DEG C, moving the rat cage to the low temperature environment of 4 DEG C, and carrying out auxiliary cooling through the ice bag. The invention further provides the rat mild hypothermia animal model and an application thereof. The invention has the advantages as follows: the target temperature is quickly reached; the body temperature fluctuation range is small; the rat skin freezing injury is reduced; and the blood sugar is controlled to be in a normal range.

Description

technical field [0001] The invention relates to the field of animal models, in particular to a rapid and effective method for preparing a rat mild hypothermia animal model. Background technique [0002] Mild hypothermia has been used as a neuroprotective therapy for more than a century and a half. Internationally, low temperature is divided into mild hypothermia: 33~36℃; moderate hypothermia: 28~32℃; deep hypothermia: 20~27℃; profound hypothermia ): below 19°C; ultra profound hypothermia: <5°C. The former two are called sub-low temperature. [0003] There are many animal experiments using rodents to study mild hypothermia therapy. The methods of implementation, maintenance and temperature detection of mild hypothermia in such experiments are summarized and analyzed as follows: [0004] 1) Put the animals in a low temperature environment of 4 degrees Celsius. Using a low temperature environment of 4 degrees Celsius alone can induce mild hypothermia in rats, but it take...
