Benefits of technology

This patented technology allows us to study how different environments can affect the appearance or severity of skin rashes caused by lupus erythematosum (Sever) virus. By studying these conditions we found correlations between specific mutation patterns on DNA from individuals with this disease and other Chinese populations who are affected negatively. These findings help identify environmental factors like sunlight exposure during childhood which could be associated with certain forms of arthritis such as polyarthrosis vulgaris).

Problems solved by technology

This patented technical problem addressed in the patents relates to studying how different individuals develop various types of diseases such as scleroderma caused by bacteria called Lyme Disease or related conditions like Autoimmunity Syndrome.

Embodiment

[0016] 1. Research object

[0017] 286 SLE patients (male 26 persons, 260 females, median age 29 years, age range 12-55 years) and 352 ethnically matched healthy controls (30 males, 322 females, median age 28 years, age range 17- 46 years old) participated in the study. All the healthy control group had no family history of SLE and related clinical symptoms. This study was approved by the Ethics Committee of Shenzhen Hospital and informed consent was obtained from all participants.

[0018] 2. Genotyping

[0019] Using Innogent's whole genome DNA extraction kit (Innogent, China), the collected peripheral blood genomic DNA was extracted according to the instructions of the kit. The genotype of the samples was identified by the high-resolution melting curve method of unlabeled probes. The method is briefly described as follows: first, asymmetric PCR was carried out, and the 20 mL system contained 20 ng of genomic DNA template, 1′PCR buffer solution (Takara, Japan), 200 mM d...

Abstract

The invention discloses a method for determining correlation between single nucleotide polymorphisms rs4840568 and rs13277113 of Han people in Chinese Mainland and systemic lupus erythematosus (SLE). The method comprises that through a non-labeled probe high-resolution solubility curve method, a SLE patient is subjected to genetic typing so that the correlation between single nucleotide polymorphisms rs4840568 and rs13277113 of Han people in Chinese Mainland and SLE is determined. At present, it is proved that environmental factors and gene factors produce important effects in SLE generation and development and thus a research result obtained by the method provides a theoretical basis for in-depth knowledge of SLE diseases.
