Problems solved by technology

In contrast, the sensitivity of colloidal gold technology is worse than that of ELISA, which can only reach 50ng / ml. It is almost impossible to detect trace amounts of biological toxins in seafood. In addition, the current domestic colloidal gold technology Still in its infancy, basically unable to quantify toxin detection

Embodiment 1

[0125] The preparation process of this embodiment is shown in figure 1 .

[0126] 1. Extraction and purification of toxins

[0127] 1.1 Extraction

[0128] Weigh the sample, place it in a clean centrifuge tube, add 80% (volume) methanol aqueous solution, preferably the mass volume ratio of the sample to the methanol aqueous solution is 2g:10mL.

[0129] After ultrasonic extraction for 5 minutes, centrifuge at 6000r / min for 10 minutes, and repeat the extraction once.

[0130] Combine the supernatants, and dilute with 80% (volume) methanol aqueous solution to obtain a constant volume solution. Preferably, the mass-to-volume ratio of the sample to the constant volume solution is 2 g:25 mL.

[0131] Take 1 / 2 of the constant volume solution in another centrifuge tube, add n-hexane, vortex mix, separate layers, discard the n-hexane layer, preferably the mass volume ratio of the sample to the n-hexane is 2g : 5mL.

[0132] Add water and chloroform, vortex mix, layer, absorb the ...

Abstract

The invention discloses a method for rapidly detecting shellfish toxin. The method comprises the following steps: (1) extracting and purifying shellfish toxin of a sample; (2) identifying the shellfish toxin; (3) preparing a shellfish toxin-BSA coupling antigen; (4) preparing and purifying a poisoning monoclonal antibody; (5) preparing immunomagnetic microspheres; (6) preparing a glucan-antibody-fluorescein mixed marker; (7) purifying the mixed marker; and (8) preparing a fluorescence immune chromatography test strip. The method is high in specificity and low in detection limit and can realize quantitative determination of antigen components.

Description

technical field [0001] The invention relates to a detection method, in particular to a method for detection of shellfish toxin. Background technique [0002] Shellfish toxins are marine natural organic matter, and its formation is closely related to toxic algae blooms in the ocean. Harmful red tides have become a worldwide problem, and the range of occurrence is still expanding, and the types of harmful algae that can form red tides are also increasing. According to literature reports, there are about 184,267 species of microalgae that can form red tides, including about 60 to 78 species of toxic microalgae. The toxins produced by toxic algae often enter the shellfish through the food chain, so these toxins are usually called shellfish poisoning. People eat shellfish with shellfish poisoning and cause food poisoning. The pollution of shellfish toxins in the four seas of my country is very serious. The survey found that there is extensive shellfish toxin pollution in all m...
