Problems solved by technology

[0003] In fact, the effectiveness and practicability of the patented invention of "sex-selective insemination of mammals with a small number of sperm cells" cannot be generalized, especially when evaluating the effect of sexually controlled cold mating of multiparous dairy cows in an all-round way, the satisfaction level is low. not optimistic

Embodiment 3

[0070] Embodiment 3 Utilize sex control semen to carry out the method (1) of cold mating of dairy cow

[0071] Semen source: Rapture, a Holstein bull, was purchased from WESTENG, Canada; chemical reagents were purchased from Sigma.

[0072] After taking the fresh semen of bulls, process the fresh bovine semen with the semen protectant prepared in Example 1. The method for treating the semen with the semen protectant is: add the semen protectant of 0.25 times the volume of bovine semen to the bovine semen.

[0073] Stain the treated bovine semen fluorescent dye Hoechst33342, and use a conventional cell sorter (using Moflo high-speed flow cytometry sorter) to sort dairy cow sperm cells; Its quality inspection: the number of sperm moving in a straight line should be more than 50%; the rate of deformed sperm should not be higher than 15%; the acrosome integrity rate should be higher than 40%; About 2 million sperm counts per milliliter were packed into tubes, cryogenically frozen...

Abstract

The invention provides a method for inseminating multiparous cows by using frozen sex-controlled semen. The method comprises the following steps that (1) bull semen is collected and processed through semen protective agents; (2) X sperms are sorted out and frozen at low temperature; (3) artificial insemination samples of the multiparous cows are manufactured, and each sample which is 0.25ml in volume comprises 2 millions-4.2 millions of sperms; (4) at least one part of each artificial insemination sample is injected into each rutting multiparous cow, and therefore at least one spawn inside each multiparous cow is fertilized; cow offspring are bred. The method enables the conception rate of the multiparous cows to be more than 10% higher that in a conventional method; the method achieves convenient operation and low cost, effectively increases the conception rate and the offspring female rate of the multiparous cows, and is suitable for breeding of the multiparous cows with the frozen semen.

Description

technical field [0001] The invention relates to a method for cold mating of dairy cows, in particular to a method for cold mating of multipartum dairy cows by using sexually controlled semen. Background technique [0002] Solving the problem of good breeding of dairy cows is a crucial link in the dairy industry and the key to the success or failure of cattle-raising enterprises. The reason why it is more difficult to reproduce dairy cows than young cows is that there are many influencing factors in dairy cows, such as milk production, uterine status, ovarian status, cow body condition, etc. As far as the cold mating of dairy cows is concerned, as early as the 1970s, a large number of studies focused on the relationship between the number of imported sperm and the conception rate, and it was concluded that the number of imported sperm was between 300,000 and 14 million, and the number of imported sperm and conception rate rate is positively correlated. The purpose of these ...
