Problems solved by technology

Serum can oxidize sulfhydryl groups. In addition to the contribution of resting sulfhydryl oxidase, there may be other macromolecules and small molecules that promote the oxidation of sulfhydryl groups. Dithiothreitol, dithiodinitrobenzoic acid, and guanidine hydrochloride can be used as the main reagents. The colorimetric method used to detect the total activity of sulfhydryl oxides in serum, but there is no report of using this detection method to diagnose diseases

Embodiment Construction

[0020] The present invention will be further described through the embodiments below in conjunction with the accompanying drawings.

[0021] Embodiments of the present invention include measuring the specific activity of resting thiol oxidase in serum by fluorescence quantitative method, measuring the total activity of oxidized sulfhydryl group in serum by using colorimetric quantitative method, and measuring albumin, globulin, alanine aminotransferase, alanine aminotransferase, Aspartate aminotransferase and alpha-fetoprotein; and receiver operating characteristic curve (ROC curve) analysis method was used to test the effect of serum indexes in diagnosing liver disease.

[0022] The population of the serum tested in this embodiment includes 269 chronic liver disease patients (including 123 liver cancer patients, 100 hepatitis B patients and 53 liver cirrhosis patients), 548 cancer patients (including 101 lung cancer patients, 91 gastric cancer patients, 86 colon cancer patien...

Abstract

The invention relates to a method for diagnosing the liver disease by utilizing the property of the serum capable of oxidizing sulfhydryls. The property of the serum capable of oxidizing the sulfhydryls is commonly contributed by resting sulfhydryl oxidase and other macromolecules or micromolecules in the serum. The specific activity of the resting sulfhydryl oxidase in the serum can be detected by a fluorescence method adopting Amplex UltraRed, horseradish peroxidase, dithiothreitol and tween as main reagents. The total activity of the oxidized sulfhydryls in the serum can be detected by a colorimetric method adopting dithiothreitol, dithiobisnitrobenzoic acid and guanidine hydrochloride as main reagents. The method and the kit show that the specific activity of the resting sulfhydryl oxidase in the serum and the total activity of the oxidized sulfhydryls in the serum can be used for diagnosing the liver disease, wherein the diagnosis effect of the total activity of the oxidized sulfhydryls in the serum is better. The invention also relates to a kit for diagnosing the liver disease. The kit adopts phosphate buffer solution, the Amplex UltraRed, the horseradish peroxidase, the dithiothreitol and the tween as the main reagents or adopts the guanidine hydrochloride, the dithiobisnitrobenzoic acid and the dithiothreitol as the main reagents.

Description

technical field [0001] The present invention relates to a method for diagnosing liver disease. The method is carried out by using the properties of serum thiol oxidized; in particular, it relates to using the specific activity of serum resting thiol oxidase or the total activity of serum oxidized thiol as biological indicators to diagnose liver disease; the present invention also It relates to a reagent kit for diagnosing liver diseases. Background technique [0002] Common liver diseases include hepatitis, cirrhosis, and primary liver cancer. There are many serum indicators for diagnosing liver disease. Alanine aminotransferase, aspartate aminotransferase, alpha-fetoprotein, and ratio of albumin to globulin are commonly used diagnostic indicators. The sensitivity and specificity of these indicators have limitations, and new serum indicators are needed to improve the diagnostic effect. [0003] Resting sulfhydryl oxidase catalyzes the formation of disulfide bonds from sulf...
