Embodiment 1

[0016] Embodiment 1: a method for inducing eosinophila somatic embryos, its implementation steps include:

[0017] (1) Selection of explants: take the flower branch of Eelium eosinophila as the explant donor, take the healthy young spathe inflorescence, scrape off the attached organisms on the surface, wash it in sea water and transfer it to the ultra-clean workbench Inside, soak in 10% hydrogen peroxide solution for 15-20min, rinse with sterilized sea water for 3 times, peel off the bract leaves outside the spathe inflorescence to obtain a sterile inflorescence;

[0018] (2) Induction culture of somatic embryos: Cut the sterile inflorescences into 3-5mm long explant fragments, inoculate them in the induction medium, and cultivate them at 16°C, under dark conditions and under 1000lux light conditions, every half a month Transfer once. After 17 days, small protrusions could be seen on the inflorescence, and after about 30 days, these small protrusions could be seen to develop ...

Abstract

The present invention relates to a method for inducing somatic embryos of Zinaria chinensis. The inflorescences of spaghetti algae are used as explants, and the surface-sterilized inflorescences are cut into 3-5mm long fragments and inoculated into somatic cells. On the embryo induction medium, conduct induction culture at 15-25°C, dark or weak light conditions, and transfer once every half month. After 15-25 days of culture, the tissue on the inflorescence can be seen to germinate, produce small protrusions, and gradually develop into somatic embryos. No callus phase was experienced during this period. The somatic embryo induction medium is a solid medium with MS and N6 medium as the base medium, supplemented with organic additives, carbon sources, plant hormones (auuxin, or auxin and cytokinin) and sea salt. The method of the present invention can directly induce the somatic embryos of Zinaria spp. It provided important technical support and laid the foundation for the success of seagrass tissue culture.

Description

technical field [0001] The invention belongs to the field of plant tissue culture research, and the invention relates to a method for inducing somatic embryos of eosinopsis. Background technique [0002] Eelweed ( Zostera marina L.) is the most widely distributed seagrass in the temperate seas of the northern hemisphere, and it is also the dominant species in seagrass farms in northern my country (Shandong, Hebei and Liaoning). It has extremely important ecological service functions and economic value. However, in the past hundred years, the global eelgrass seagrass ecosystem has been seriously damaged, and the area has shrunk sharply, even worse in my country. Therefore, in recent years, the restoration of eelgrass seagrass has become an indispensable item in the restoration of marine ecosystems in northern my country. Contents, a large number of seedlings are needed in the grassland restoration, and using plant tissue culture technology to cultivate sterile seedlings indo...
