Embodiment 1

[0019] 1. Establishment of sterile culture system

[0020] Cut off the germinated shoots of Lonicera early-flowering that year in early May, rinse with running water for 1 hour, then trim off the leaves, add detergent to soak for 10 minutes, and rinse with running water. Finally, put them on the ultra-clean workbench and use the following three treatments to screen for suitable sterilization methods.

[0021] The first one is soaking in 0.1% mercuric chloride for 5 minutes and washing with sterile water for 6 times;

[0022] The second is to soak in 0.1% mercuric chloride for 7 minutes and rinse with sterile water for 6 times;

[0023] The third is to soak in 2.0% sodium hypochlorite for 20 minutes, and rinse with sterile water for 3 times.

[0024] Inoculate the stem section with buds about 1 cm long into MS blank medium, inoculate 60 bottles for each treatment, and repeat 3 times. After culturing for 30 days, the contamination rate and survival rate were counted. Contami...

Abstract

The invention belongs to the field of plant tissue culture, and provides an in-vitro rapid propagation method for lonicera praeflorens. The method comprises the steps that tender stems of lonicera praeflorens are used as an explant, an MS is used as a minimal medium, plant growth regulators 6-BA and NAA are added for inducing axillary buds to germinate and grow; then, subculture multiplication, rooting induction and acclimatization and transplanting are performed, and a large number of regeneration plants can be obtained within a short period of time. The effective way is provided for rapid propagation of high-quality seedlings of lonicera praeflorens, the growth coefficient is high, the induction rooting rate is high, and transplanting and survival are easy. The requirement for landscaping in early spring in the Northeast China region can be met to a great extent, and a foundation is laid for further comprehensive development and utilization.

Description

technical field [0001] The invention belongs to the field of plant tissue culture, and in particular relates to an in vitro rapid propagation method of honeysuckle early flowering. Background technique [0002] Lonicera praeflorens Batalin is a deciduous shrub of the genus Lonicera in the family Caprifoliaceae. It is mainly distributed in the southeast of the three northeastern provinces of my country, and it is also distributed in Russia (Far East), North Korea, and Japan. Early-flowering honeysuckle blooms before the leaves in mid-to-late April, and they are born in pairs on the main peduncle. They are lavender and show elegant and unique charm in early spring. This tree species has lush branches and leaves, early flowering, and early fruit ripening. The green leaves set off the red fruit, which is very beautiful. It can make up for the depression in early spring in Northeast China, prolong the greening time of the city, and enrich the greening landscape in early spring....
