The method includes the steps of sufficiently grinding dried stigmas of crocus sativus through liquid nitrogen, adding 4*CTAB and phenol-chloroform of the same volume, conducting concentration, adding pre-cooling anhydrous ethanol and NaAc for sedimentation, conducting washing absolute ethyl alcohol, and storing the samples in a refrigerator with the temperature of -20 DEG C for standby application, wherein the volume of the pre-cooling anhydrous ethanol is two times of the volume of phenol-chloroform, the volume of NaAc is 20% of the volume of phenol-chloroform, and the volume of absolute ethyl alcohol is 2.5 times of the volume of phenol-chloroform.
