Abstract

The invention discloses a tissue culture method for ipomoea holubii, which particularly includes the steps of: 1) preparation of a culture medium which includes components of a basic culture medium and culture mediums in all steps of tissue culture; 2) selection and disinfection of an explant; 3) induction of axillary buds; 4) proliferation of adventitious buds; and 5) rooting and transplanting. The method has the following advantageous effects: through plant tissue cultivation, induction of axillary buds and proliferation of adventitious buds clusters are carried out to stem fragments of the ipomoea holubii, so that a large quantity of high-quality seedlings having consistent genetic characters can be obtained within short time. The method also overcomes defects of scarcity of female parents and low speed of common reproductive methods, and also has positive significance on protection of germplasm resources of wild ipomoea holubii and promotion of seedling production in large scale.
