[0027] Detection of profenofos using the helper chain as a probe

[0028] Materials / Reagents: Terbium trichloride was purchased from Sigma-Aldrich Reagent Company; Tris (Tris) was purchased from Sinopharm Chemicals Co., Ltd.; nucleotide chains were synthesized by Sangon Bio (Shanghai) Reagent Company ;

[0029] Method: Take two 2mL centrifuge tubes and label them as a and b respectively. In tube a, add 50 μL of 10 μM terbium, 50 μL of 4 μM helper chain, and 400 μL of Tris-hydrochloric acid buffer solution with pH 7.9, and shake fully with a vortex for 14 minutes; add 4 μM 50 μL of helper chain, 50 μL of 6 μM nucleic acid aptamer, fully shaken for 16 minutes, then added 350 μL of Tris-hydrochloric acid buffer solution with pH 7.9, and 50 μL of 10 μM terbium, and the mixed solution was reacted at room temperature for 14 minutes, and its fluorescence value was compared with a fluorescence spectrophotometer. Fluorescence curves recorded twice.

Abstract

The invention provides a profenofos fluorescence detection method based on terbium and an aptamer. The profenofos fluorescence detection method is characterized in that the fluorescence effect of terbium can be enhanced through utilizing a G-enriched nucleotide single chain (a helping chain), and a specific recognition effect of a nucleic acid aptamer on a target object; the profenofos fluorescence detection method comprises the following steps: observing a reaction of the helping chain, the nucleic acid aptamer, the terbium and a profenofos system through determining a fluorescence spectral pattern; and detecting the selectivity of a non-labeled nucleic acid aptamer sensor on the profenofos through determining the fluorescence spectral pattern, and determining the profenofos in an actual sample through fluorescence detection. The method provided by the invention has the advantages of high specificity, rapidness, high sensitivity and simplicity and convenient operation, and lays a foundation for production in the future.

Description

technical field [0001] The invention discloses a profenofos fluorescence detection method based on terbium and an aptamer, which belongs to the technical field of analytical chemistry. Background technique [0002] Profenofos is a moderately toxic non-systemic broad-spectrum organophosphorus insecticide, which has contact and stomach poisoning effects, and has a good control effect on pests on cotton, fruit trees, rice, vegetables and other crops. The use amount is increasing day by day, and the pollution caused by the use on vegetables cannot be ignored. Although a small amount of profenofos residues will not cause immediate and direct poisoning to the human body, the molecular structure of profenofos is relatively stable, and it is difficult to be metabolized and excreted in the living body. Long-term consumption of this contaminated vegetable can cause Cancer, infertility, endocrine disorders and other diseases. Eating a lot of vegetables that exceed the standard will h...
