Problems solved by technology

Different types of idiopathic inflammatory myopathy can involve different target organs such as skin, lung, and muscle, so the clinical manifestations are more complicated, and it is difficult to achieve early diagnosis and early treatment. Malnourished patients have similar pathological changes, so some patients are easy to be misdiagnosed

Embodiment 1

[0028] Example 1 High-density protein chip screening PM / DM related autoantibodies

[0029] The N-terminal of all protein antigens on the high-density protein chip has a GST tag for protein purification. First, use rabbit anti-GST monoclonal antibody to hybridize with chip protein to verify chip quality. Then, 40 PM / DM patient sera, 30 autoimmune disease control sera, and 20 healthy control sera were selected for hybridization with 90 high-density protein chips, and candidate PM / DM-related autoantigens were identified through signal acquisition and data analysis.

[0030] 1.1 Extraction of high-density protein chip scanning data

[0031] The GenePix 4000B chip scanner was used to scan the high-density protein chip, and after obtaining the grayscale image in TIFF format, according to the operation manual of the GenePix Pro 6.0 software, and according to the high-density protein chip loading matrix parameters, the analysis software automatically completed each protein point. Se...

Embodiment 2

[0052] Example 2 Construction of PM / DM Related Autoantibody Target Antigen Protein Chip and Serum Screening Verification

[0053] Due to differences in genetic background and environmental factors in patients with autoimmune diseases, the types of autoantibodies produced by different patients with the same autoimmune disease are quite different. In order to confirm whether the PM / DM-related autoantibodies screened from small samples by high-density protein chips have clinical significance and application value, as well as the sensitivity and specificity of these autoantibodies to the diagnosis of PM / DM patients, this study will use Example 1 PM / DM-related autoantibody target antigens screened by high-density protein chips were prepared into PM / DM-related autoantibody target antigen protein chips for clinical expansion sample verification. Serum samples used for PM / DM-related autoantibody target antigen protein chips for clinical large-scale validation testing included 397 PM / D...

Abstract

The invention discloses the use of signal peptide recognition particle 19, namely SRP19, or its fragments in the preparation of reagents for diagnosing idiopathic inflammatory myopathy. The invention adopts high-density protein chip technology to screen PM / DM related autoantibody target antigens, and determines 91 PM / DM related autoantibodies. 91 PM / DM-related autoantibodies corresponding to target antigens were used to construct and prepare a PM / DM-related autoantibody target antigen chip for clinical expansion sample verification. The results showed that the positive rate of anti-SRP19 antibody in the PM group was 19.00%, which was higher than the positive rate of all control groups (2.67% (P=3.26×10 ‑7 ), the result difference was significant; the positive rate in the DM group was 7.41%, which was higher than the positive rate of all the control groups 2.67% (P=0.80×10 ‑2 ), the result difference was significant; the positive rate in the PM / DM group was 10.33%, which was higher than the positive rate of 2.67% in all control groups (P=0.09×10 ‑3 ), the difference in the results is significant. It shows that anti-SRP19 antibody can be used as a diagnostic and detection marker for idiopathic inflammatory myopathy.

Description

technical field [0001] The invention belongs to the field of biological detection, and in particular relates to a biomarker for detection of polymyositis and / or dermatomyositis and application thereof. Background technique [0002] Idiopathic inflammatory myopathies (IIMs) are heterogeneous diseases characterized by proximal symmetrical muscle weakness and multiple organ involvement, mainly including polymyositis (PM), dermatomyositis ( dermatomyositis (DM), immune-mediated necrotizing myopathy (IMNM), sporadic inclusion body myositis (sIBM), juvenile idiopathic myositis (JIM), etc. clinical subtype. PM / DM is an autoimmune disease of striated muscle nonsuppurative inflammatory myopathy. Its clinical features are elevated serum muscle enzymes, myogenic EMG changes, inflammation and degenerative changes in muscle tissues such as limb girdle muscles, neck muscles, and pharyngeal muscles, resulting in symmetrical muscle weakness and a certain degree of muscle atrophy. Involve...
