Embodiment 1

[0018] The extraction method of endophytic fungus in the tall fescue leaf of the present embodiment, comprises the following steps,

[0019] (1) take off the blade of tall fescue plant, place 15 hours in aseptic operation table;

[0020] (2) Put the leaves into 75% alcohol solution to wash for 30 seconds, then put them into 0.5% sodium hypochlorite solution to wash for 10 seconds, wash them with clear water twice, and put them on sterile paper to dry for later use;

[0021] (3) the leaves obtained in step (2) are punched with a puncher that has been sterilized in advance by high temperature and high pressure, and the circular leaves obtained are placed on the culture medium of a glass dish with a diameter of 9 cm;

[0022] (4) Adopt preservative film to seal the glass dish, and fuse the sealing strip by an alcohol lamp;

[0023] (5) Place the glass dish in a dark room at 25 degrees Celsius for 10 days to obtain colonies of endophytic fungi;

Abstract

The invention discloses an extraction method for endophytic fungi in festuca elata Keng ex E.Alexeev leaves. The method includes taking a festuca elata Keng ex E.Alexeev leaf, and placing the leaf ina sterile console for 15-20 hours; putting the leaf in an alcohol solution with 75% to clean for 30-45 sec, then putting the leaf in a sodium hypochlorite solution with 0.5% to clean for 10-15 sec, and putting the leaf on a sterile paper to dry for later use after cleaning the leaf for 2-3 times with clear water; performing hole punching on the leaf by using a hole punch, and placing the obtainedcircular leaf on a culture medium of a glass dish; sealing the glass dish, and performing fusion on a sealing strip through an alcohol burner; placing the glass dish in a darkroom under an environmentof 25 DEG C to culture for 10-15 days so that endophytic fungal colonies can be obtained; and picking out the colonies by using an inoculating needle to perform individual culturing so that endophytic fungus strains can be obtained. Compared with the prior art, the method has characteristics of high fungi extraction efficiency, high extraction survival rate and low cost.

Description

technical field [0001] The invention relates to the field of fungi extraction, in particular to a method for extracting endophytic fungi from leaves of tall fescue. Background technique [0002] Scientific name of tall fescue: Festuca elata Keng ex E. Alexeev, also known as: Fescue, belongs to Gramineae, Gramineae is a perennial ground cover plant. Classified by function: divided into lawn type (viewing), grass type (as pasture for raising livestock). It likes cold, humid and warm climates, and grows well in fine loamy soils that are fertile, moist, rich in organic matter, and have a pH of 4.7 to 8.6. It is widely used in sports field turf and protective turf. Feed livestock as pasture. [0003] At present, endophytic fungi have been found in tall fescue, but there is no mature method for their extraction. Contents of the invention [0004] Purpose of the invention: Aiming at the deficiencies and defects of the prior art, the present invention provides a method for ext...
