[0059] Example 1: In vitro biofilm studies with thermolysin and collagenase

[0060] In vitro assays were performed to demonstrate the bacterial biofilm reducing capacity of thermolysin. In this assay, S. aureus ATCC 6538 was suspended in growth medium of tryptic soy broth supplemented with 0.25% glucose for optimal bacterial biofilm formation. The suspension was transferred to the wells of a sterile 96-well culture plate and incubated at 37°C for 22 hours with one medium change. After the bacterial biofilm was formed, the growth medium was replaced with a thermolysin solution dissolved in the growth medium at different concentrations and a collagenase solution dissolved in the growth medium at the same concentration (enzyme concentration was 0.000001 mg / mL, 0.00001mg / mL, 0.0001mg / mL, 0.001mg / mL, 0.01mg / mL, 0.1mg / mL, 1.0mg / mL and 10mg / mL). Methods are disclosed for the reduction or elimination of bacterial biofilms on biological and non-biological surfaces, as well as methods for the treatment of wounds, skin lesions, mucous membrane lesions, and other biological surfaces infected or contaminated with bacterial biofilms using compositions comprising thermolysin.
