Embodiment

[0030] This embodiment provides a method for regulating the aggregation state of human insulin, including the following steps:

[0031] 1. Mix the fast green powder and deionized water ultrasonically and shake for 1 minute under 200w conditions to form a 1 mM fast green solution;

[0032] 2. Mix hIAPP with phosphate buffered saline solution to form a 500μM protein solution;

[0033] 3. Add deionized water to the protein solution and dilute with shaking to reduce the concentration of the protein solution to 50μM;

[0034] 4. Divide the 1mM fast green solution into three parts, and then dilute them into 8μM, 80μM and 800μM fast green solutions;

[0035] 5. Take out 40 μL from the protein solution, and then add 40 μL of deionized water to form a blank group with a hIAPP concentration of 25 μM;

[0036] 6. Take out 40μL from the protein solution, and then add 40μL of 8μM fast green solution to form sample 1 with hIAPP concentration of 25μM and fast green concentration of 4μM;

[0037] 7. Take...

Abstract

The invention discloses a method for regulating and controlling the aggregation state of human islet protein, which comprises the following steps: mixing hIAPP with a phosphate buffer salt solution toform a protein solution, mixing fast green with deionized water to form a fast green solution, and mixing the fast green solution with the protein solution to form a mixed solution. According to themethod, the aggregation degree of the hIAPP is regulated and controlled through fast green, and the aggregation of hIAPP can be effectively inhibited or promoted. The low-concentration fast green canblock the formation of the hIAPP dimer, so that the survival rate of pancreatic islet beta-cells is effectively increased, the high-concentration fast green can effectively promote the aggregation ofhIAPP, and the aggregation degree is effectively increased compared with that when fast green is not added, so that the effect of increasing the aggregation degree of hIAPP is achieved. In addition, under the condition of high-concentration fast green, the aggregation speed of hIAPP is increased, and the existence time of hIAPP toxic intermediates (dimers and the like) is shortened, so that the survival rate of pancreatic islet beta-cells is increased from another perspective. Therefore, fast green can also be used as a medicine for preventing and treating type 2 diabetes mellitus.

Description

【Technical Field】 [0001] The invention relates to a method for regulating the aggregation state of human islet protein and a treatment and preventive medicine for type 2 diabetes, belonging to the field of biological protein regulation. 【Background technique】 [0002] Amyloid aggregates (Amyloid) is an insoluble fibrous protein. If it accumulates abnormally in organs, it will cause amyloidosis. In many neurological diseases, such as Alzheimer's disease, Parkinson's disease and type II diabetes, a large amount of amyloid fiber accumulation can be observed in the system. Most scholars believe that it may cause degeneration or dysfunction of the brain or other related organs. The production of amyloid aggregates undergoes a process from monomers to oligomers. This process is called the nucleation period. Then the oligomers aggregate to form precursor fibers, and then rapidly form fibers, finally reaching stable mature fibers. Since the whole process is irreversible, it becomes ex...
