Abstract

The invention discloses a method for preparing Thymalfasin. The method comprises the following steps: (1) preparing a Fmoc-Asp-resin complex; (2) preparing a protected Thymalfasin-resin complex; (3) removing a lateral-chain protective group by employing a cutting reagent, splitting Thymalfasin from resin, and carrying out ethyl ether precipitation, so as to obtain a Thymalfasin crude product; and (4) purifying the crude product by employing high-performance liquid chromatography, thereby obtaining refined Thymalfasin. The method for preparing the Thymalfasin, provided by the invention, is simple, convenient and efficient, and the problem that Thymalfasin preparation processes are tedious and are low in efficiency and high in cost at present is solved.

Description

technical field [0001] The invention belongs to the field of polypeptide chemistry, and in particular relates to a new preparation method of thymus method. Background technique [0002] Thymofasin consists of 28 amino acids, the N-terminal contains acetylation structure, the molecular weight is 3108, and the primary structure is Ac-Ser 1 -Asp 2 -Ala 3 -Ala 4 -Val 5 -Asp 6 -Thr 7 -Ser 8 -Ser 9 -Glu 10 -Ile 11 -Thr 12 -Thr 13 -Lys 14 -Asp 15 -Leu 16 -Lys 17 -Glu 18 -Lys 19 -Lys 20 -Glu 21 -Val 22 -Val 23 -Glu 24 -Glu 25 -Ala 26 -Glu 27 -Asn 28 -OH. Thymalfasin is a regulator of biological responses, an immune enhancer for T lymphocytes, it can promote the maturation and differentiation of T cells, and can promote mature T cells and NK cells to secrete a variety of lymphokines (IL -2, INF-α, INF-γ, etc.), but also promote the production of high-affinity interleukin-2 receptor (IL-ZR). In addition, thymofasin has a direct inhibitory or killing effect...
