A third exon in the IGFBP7 gene is knocked out from the mouse model, an F0 chimeric mouse is constructed through construction of a targeting vector and ES targeting in sequence, the F0 chimeric mouse and a tissue-specific Cre tool mouse are hybridized to obtain an F1-generation knockout heterozygote, and finally, the F1-generation heterozygote is subjected to inbreeding to obtain an IGFBP7 gene knockout homozygote mouse.
