The method comprises the following steps: 1) extracting total alkaloids; 2) separating the picrinine to obtain Fr-A containing the picrinine, a light yellow solid Fr-B, a grey white solid Fr-C and yellowish acicular crystals Fr-D, preparing achromatic acicular crystals from the Fr-D part, and recrystallizing by acetone to obtain achromatic acicular crystals, namely a picrinine pure product which is the picrinine reference substance. The reference substance is in accordance with the technical requirement on the chemical reference substance for traditional Chinese medicinal quality standard for content measurement. The picrinine reference substance has good stability, and can be used for content analysis work.
