Embodiment Construction

[0041] The zmERF12 promoter was cloned into the transformation vector, followed by a Tnos terminator to construct a transformation vector. The transformed target gene was cloned between the zmERF12 promoter and the Tnos terminator in the correct direction to construct a transformation vector. Then, the target gene is transformed into recipient cells by gene gun bombardment method or Agrobacterium-mediated method, and transgenic plants are obtained by technical means such as tissue culture. Under waterlogging and flooding conditions, the zmERF12 promoter can promote the massive expression of the target gene in the root of the plant.

Abstract

The invention relates to a maize flooding stress response zmERF12 gene promoter and is characterized by disclosing a DNA sequence of the zmERF12 gene promoter. The promoter is characterized by comprising cis-elements, namely two G-boxes, five ABREs, a CCAAT-box, four CGTCA-motifs, a GARE-motif, a GC-motif, a TCA-element, a TGA-element and an as1, wherein the cis-elements are related to plant hormones and anaerobic stress responses. The promoter is a tissue-specific promoter only expressed in the root system and an inductive promoter efficiently expressed under flooding induction. The insect-resistant, disease-resistant and adversity-resistant capabilities of the roots of transgenic plants can be improved by the promoter under flooding and waterlogging conditions. Meanwhile, the promoter is only expressed in the roots so that food safety problems cannot occur when the overground parts of the transgenic plants are eaten. The promoter has an excellent application prospect in the waterlogging-resistant genetic improvement of xerophilous plants.

Description

Technical field: [0001] The invention relates to a zmERF12 gene promoter responding to water stress in maize, belonging to the technical field of plant genetic engineering. Background technique: [0002] In the normal growth environment of southern corn, due to seasonal rainfall, it often suffers continuous spring rains at the seedling stage, and often encounters plum rains at the flowering and filling stages. Under long-term rainy conditions, poor irrigation and drainage systems and rising groundwater levels cause soil waterlogging, which keeps the corn root system in a low-oxygen state for a long time, resulting in a 15%-25% drop in yield, sometimes even up to 50%. Waterlogging has become an important factor restricting the stable and high yield of maize in southern China. Therefore, understanding the tolerance / sensitivity response of maize to waterlogging and identifying important waterlogging tolerance genes are very important for elucidating the molecular biological me...
