Problems solved by technology

[0004] At present, forensic workers at home and abroad have used biophysics, biochemistry, immunohistochemistry, molecular biology and other methods and technologies, and based on the content of material components and the postmortem sequence of physical and chemical properties caused by autolysis and corruption of biological tissues However, due to the comprehensive influence of autolysis, decay, and various factors inside and outside the body, existing methods for inferring PMI have technical bottlenecks such as single indicators, short window periods, and poor accuracy.

[0020] Select 10 4-month-old healthy domestic pigs with an average weight of about 150kg. After slaughtering, extract the brain tissue of each pig and place them in an artificial climate box with a constant temperature of 25°C, a constant humidity of 100%, and a daytime light of 12 hours to allow them to autolyze and rot.

[0021] According to the standardization of sampling "six determinations" method: 1. Timing: after death, 11 time groups of 0h, 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, and 120h were respectively collected in the brains of 5 pigs. Sampling on the tissue sample; 2. Positioning: take the frontal lobe area of the brain tissue, and take a sample containing a ratio of cortex to white matter of 1:1; 3. Quantification: prepare 80% alcohol fixative solution according to 1:4 distilled water / absolute ethanol; 4. Shape-setting: 10g of specimens of the same size are taken at the designed time points and locations; 5. Fixed ratio: the mass-volume ratio of brain tis...

Abstract

The invention relates to a method for fixing and extracting a biological tissue by using immobile liquid to deduce the death time of the biological tissue. By adopting the method, kinds of detection methods are applied to analyze on the basis of the principle that immobile liquid can quickly terminate self-digestion and decay change processes of an in vitro biological tissue, so as to extract a material composition in the biological tissue and achieve dynamic balance within a certain period of time; finally, the postmortem interval (PMI) is indirectly deduced according to the scheduling rule of occurrence and development of self-digestion and decay of the biological tissue. The method comprises the steps: sampling 'six-fixing' method, drafting of a 'window phase' standard curve, and deducing of the PMI. By adopting the method disclosed by the invention, the in vitro biological tissue can be effectively prevented from continuing to generate self-digestion and decay between the processes of drawing materials and detecting by the sampling 'six-fixing' method, so that the accuracy and the scientificity of deducing the PMI are improved.

Description

technical field [0001] The invention relates to a method for estimating the postmortem interval (PMI), in particular to a method for fixing and extracting biological tissues with a fixative to estimate the time of death. Background technique [0002] PMI refers to the time interval between the moment of death and the moment of examination. In the detection of criminal cases, PMI is of great significance to the judiciary of determining the time of the incident, identifying suspects, and delineating the scope of investigation. It also has certain significance for the handling of inheritance, insurance claims and other cases. Since the time of occurrence of many unnatural death cases is not clear, how to accurately infer PMI has always been the most important issue in the actual forensic identification work. [0003] After the death of humans or other animals, as time goes by, the corpse will undergo a series of post-mortem changes in the corresponding environment, among which...
