Obviously, the described embodiments are only some, not all, embodiments of the present invention. Based on the embodiments of the present invention, all other embodiments obtained by persons of ordinary skill in the art without creative efforts fall within the protection scope of the present invention.

[0024] A method for green fluorescent protein gene labeling of the snakehead source Aeromonas schubert, comprising the following steps:

[0025] (1) Isolation of the snakehead source Aeromonas schubert:

[0026] (2) Preparation of promoter fragments:

[0027] The PCR reaction was performed according to a conventional method. The template is the plasmid PRUAL (SEQ NO: 1), and the upstream and downstream primers are ppS and ppA, respectively. The PCR reaction syst...

Abstract

The invention provides a method for labeling aeromonas schubertii from snakehead through green fluorescent protein genes. According to the method, a pMDGFP recombinant carrier is constructed and introduced into an aeromonas schubertii virulent strain WL-2. When being observed under a fluorescence microscope, thallus can present green fluorescence. Stability tests show that the plasmid stability rate can be 100% when the WL2GFP passed to thirtieth generation. By means of the green fluorescent protein genes, a simple and visual method for researching the mutual relation between the aeromonas schubertii and a host is provided.

Description

technical field [0001] The invention relates to the field of biological monitoring, in particular to a method for marking the aeromonas schubert with a green fluorescent protein gene. Background technique [0002] Channidae (Channidae) is an important freshwater economic fish in my country. It is widely distributed in China. Its meat is tender, delicious, and has high nutritional value. Channa maculata and Channa argus Channa maculata ♀×C. argus♂ is an important cultured species in my country, and it is also an important freshwater economic fish for export in my country. With the development of high-density culture of pond snakeheads in my country, the problem of farmed snakehead fish diseases is becoming more and more serious, and the pathogens that lead to the occurrence of snakehead fish include Aeromonas hydrophila, Nocardia, Schubert Aeromonas etc. Channa viscera sarcoidosis is a new outbreak of bacterial venereal disease of Channaidae fish in recent years. The disease ...
