The method specifically comprises the following steps: (1) pretreating a sample: treating the tobacco sample, so as to obtain a solution to be detected; (2) preparing a standard solution; (3) carrying out qualitative detection on the sample: carrying out ultrahigh-performance bonded-phase chromatography detection on the solution to be detected and the standard solution separately, comparing retention time to carry out qualitative detection, and determining ingredients of the nicotine chiral isomers in the solution to be detected; (4) carrying out quantitative detection on the sample: carrying out ultrahigh-performance bonded-phase chromatography detection on the solution to be detected and the standard solution separately, and carrying out quantification by adopting an external standard method, so as to obtain the content of the nicotine chiral isomers in the solution to be detected.
