Abstract

The invention relates to a method for cryopreservation of donkey semen. The method comprises (1) preparing a base solution from 3.04 parts by mass of Tris, 1.70 parts by mass of citric acid, 1.50 parts by mass of glucose, 1.50 parts by mass of fructose, 0.06 parts by mass of penicillin, 0.10 parts by mass of streptomycin and 100 parts by mass of double distilled water and preparing a semen diluent from 85 parts by mass of the base solution and 15 parts by mass of an egg yolk solution, (2) using a rutting mare as a platform ass and collecting semen of a stud ass through a false vaginal method, wherein a motility rate is greater than 0.70, (3) taking the semen diluent and semen at 28 DEG C and carrying out dilution according to a ratio of 2: 1, (4) putting the semen diluent into a centrifuge tube, putting the centrifuge tube in a water bath at 28 DEG C and transferring the centrifuge tube into a refrigerator at 4 DEG C, and (5) in cryopreservation, carrying out uniform mixing every 24 hours and simultaneously detecting sperm viability through a microscope.
