Embodiment Construction

[0025] The present invention is further explained and illustrated by specific examples below.

[0026] In this example, 50 cockles were selected, including 28 cases in Xiapu, Fujian, and 22 cases in Yangjiang, Guangdong.

[0027] In this embodiment, the amplification primers for the mitochondrial COI gene of green cockles are screened through the following steps:

[0028] 1. Log in to the National Center for Biotechnology Information (NCBI, http: / / www.ncbi.nlm.nih.gov / ), and download 12 existing mitochondrial COI sequences of green cockles (Genbank accession numbers: KJ774469, KU341920-341927 , HQ258840-258841).

[0029] 2. Use the sequence processing software Mega6.0 to conduct multiple alignment analysis on the above sequences, and determine the relatively conserved regions at the front and back ends.

[0030] 3. After comparison, use the primer design software PrimerPremier 5 to design primers in the relatively conserved regions of 100 base pairs at the front and rear end...

Abstract

The invention discloses an amplification primer for an arca virescens reeve mitochondria COI gene. The primer sequences are COIQ-F:5'-TCATATTCGGCTAAATCTT-3' and COIQ-R:5'-CATCTTTCCACACCTCAA-3'. The amplification primer for the arca virescens reeve mitochondria COI gene is capable of effectively amplifying an arca virescens reeve COI gene fragment, the amplification length is about 600bp and the amplification efficiency can reach 94%; the amplification primer is suitable for genetic diversity research of the arca virescens reeve; and meanwhile, a foundation is laid for genetic resource conservation and utilization of the arca virescens reeve and genetic diversity evaluation.

Description

technical field [0001] The invention relates to a screening method for amplifying primers of cytochrome coxidase subunit I (COI) gene of green cockles, and belongs to the field of gene bioinformatics analysis related to fishery. Background technique [0002] Green cockles ( Barbatia virescens ) belongs to the phylum Mollusca, the class Bivalvia, the order Taxodonta, the family Arcidae, the genus Clams ( Barbatia ), mainly distributed from the south of Shengshan Mountain in Zhejiang Province to the coast of Hainan, and also distributed in Japan, Vietnam, the Philippines and other countries. Green cockles mainly inhabit shallow water areas tens of meters deep in the intertidal zone, and attach to the crevices of rocks and reefs with foot silk. As an economic species in the cockle family shellfish, the green clam has delicious meat and rich nutrition, and has great potential for breeding and development. As a filter-feeding species in coastal ecosystems, cockles play an imp...
