Sequences of the primers are as follows: the sequence of an upstream primer is shown in SEQ ID NO.1, namely, COIF: 5'-CAACGTAATTGTCACTGCCCACG-3'; and the sequence of a downstream primer is shown in SEQ ID NO.2, namely, COIR: 5'-GTATCTGTGATCTGCTGGTGGGA-3'. By means of the macrobrachium rosenbergii mitochondrial COI gene amplification primers, the length of an obtained macrobrachium rosenbergii mitochondrial COI gene segment can be increased to about 1300 bp, and the primers have the characters of a single band and high amplification efficiency so that the requirements of subsequent macrobrachium rosenbergii mitochondrial genome research and macrobrachium rosenbergii population genetic diversity research can be met better.
