Embodiment 1

[0027] A preparation method of honeysuckle leaf anti-inflammatory active components, the steps are as follows:

[0028] 1) Extraction of chemical components of honeysuckle leaves: About 114g of fresh honeysuckle leaves were picked and dried in a 40°C oven to constant weight to obtain about 37g of dried honeysuckle leaves, which were crushed and passed through a 60-mesh sieve. Precisely weigh 10g of dried honeysuckle leaf samples that have passed through a 60-mesh sieve and place them in a Soxhlet extractor, add 200ml of petroleum ether and carry out degreasing treatment at 80°C until the solution in the Soxhlet extractor becomes colorless. After defatting, it was air-dried, and 500ml of 80% methanol was used for reflux extraction at 80°C for 3 times, each time for 3 hours. After the extraction, the extracts were combined for 3 times to obtain a tan viscous crude extract;

[0029] 2) Dissolve the crude extract to a suspended state with secondary water ultrasonically, extract wi...

Embodiment 2

[0034] A preparation method of honeysuckle leaf anti-inflammatory active components, the steps are as follows:

[0035] 1) Extraction of chemical components of honeysuckle leaves: Precisely weigh 10g of dried honeysuckle leaf samples after passing through a 60-mesh sieve and place them in a Soxhlet extractor, add 400ml of petroleum ether for degreasing treatment at 80 °C, until the solution in the Soxhlet extractor changes. It can be colorless. After defatting, it was air-dried, and 300ml of 80% methanol was used for reflux extraction at 80°C for 3 times, each time for 3 hours. After the extraction, the extracts were combined for 3 times to obtain a tan viscous crude extract;

[0036] 2) The crude extract is ultrasonically dissolved in secondary water to a suspended state, extracted with an equal volume of n-butanol, and the extract is concentrated under reduced pressure to obtain a n-butanol phase;

[0037] 3) Separation by column chromatography: use a 1.6cm×50cm chromatogra...

Abstract

The invention relates to a preparation method of a honeysuckle leaf anti-inflammatory active component. The preparation method comprises the following steps: 1) performing petroleum ether degreasing treatment on the crushed dry honeysuckle leaves and performing reflux extraction by using methanol to obtain a viscous crude extract; 2) dissolving the viscous crude extract by using secondary water toobtain suspension, extracting by using normal butanol and performing reduced-pressure concentration to obtain a normal butanol phase; and 3) performing column chromatography isolation on the normal butanol phase, eluting by using a methanol aqueous solution of 10 percent, 20 percent, 30 percent, 40 percent, 50 percent, 60 percent and 70 percent separately, and eluting the obtained fraction by selecting a methanol aqueous solution of 50 percent. Through research on the inhibition effect of 5-LOX, research on the anti-inflammatory effect of a mouse macrophage system RAW264.7 and research on theanti-inflammatory effect of rat foot swelling, the invention discovers that the honeysuckle leaf anti-inflammatory active component (M1) has excellent anti-inflammatory activity and can provide theoretical basis for the research and development of the anti-inflammatory medicine.

Description

technical field [0001] The invention belongs to the technical fields of analytical chemistry and biomedicine, and particularly relates to an anti-inflammatory active component of honeysuckle leaves, a preparation method and an anti-inflammatory application thereof. Background technique [0002] Lonicera japonica (English name: Lonicera japonica), also known as "Honeysuckle", is a semi-evergreen vine, Lonicera genus, first recorded in "Famous Doctors". Honeysuckle is a commonly used traditional Chinese medicine with a long history. Since 1995, more than 500 kinds of Chinese medicine prescriptions containing honeysuckle have been included in the "Pharmacopoeia of the People's Republic of China". Honeysuckle leaves (Leaves of Lonicerajaponica, referred to as LLJ), as early as the Ming and Qing Dynasties, it was recorded that "the stems, leaves and flowers of honeysuckle have the same functions" and "vine leaves can be used". Honeysuckle is cold in nature and sweet in taste, an...
