The method includes: pretreating dried frog oil through segmented hydration and rehydration, defatting and deodorization, and calcium salt confinement; then adding a complex protease for segmented pulsed ultrasonic enzymatic hydrolysis; after enzymatic hydrolysis, the mixture undergoes enzyme inactivation, centrifugation to remove impurities, and dual-stage ultrafiltration at 2kDa and 500Da to collect a 0.5–2kDa frog oil polypeptide initial enrichment solution, which is then purified by macroporous adsorption resin and separated by gel chromatography, followed by freeze-drying to obtain the frog oil polypeptides. This invention, through the combination of calcium salt confinement pretreatment and segmented pulsed ultrasonic enzymatic hydrolysis, allows frog oil proteins to undergo controlled enzymatic cleavage under moderate expansion conditions, which is beneficial for improving the formation stability and enrichment consistency of the 0.5–2kDa target polypeptide component. The obtained frog oil polypeptides can serve as active components for preventing skin aging.
