Problems solved by technology

However, although these molecules do show increased acceleration in motile sperm, a significant disadvantage is premature stimulation of the acrosome reaction (AR)
This results in the sperm being unable to penetrate the oocyte once it reaches the egg

Embodiment 3

[0128] Example 3 demonstrates that a La-1-like peptide reagent comprising a peptide according to SEQ ID NO. 1 according to one embodiment of the present invention increases the fertilization rate in a mouse in vitro fertilization experiment. Fertilization rates were measured in terms of the number of mouse embryos at the two-cell stage. The results showed that in the presence of the La-1-like peptide agent, the fertilization rate was increased by about 20% compared to the control. These data also indicate that La-1-like peptide reagents are non-toxic and do not prevent embryonic development.

[0129] According to an embodiment of the present invention, said La-1-like peptide agent increases the fertilization rate in in vitro fertilization procedures, in particular ICSI. Specifically, ICSI is implemented in humans.

[0130] According to an embodiment of the present invention, said La-1-like peptide agent increases the fertilization rate in artificial insemination procedures, ...

Embodiment 4

[0132] Example 4 describes how the incubation of the peptide according to SEQ ID NO. 1 with non-human primate testicular spermatozoa leads to an increase in the non-progressive and forward motility rates of the sperm cells in the sample. The results are shown in image 3 and 4 middle. The increase in motility in these testicular spermatids was similar to what occurs during maturation that normally occurs in the epididymis.

[0133] According to an embodiment of the present invention, testicular spermatozoa that have exhibited increased motility after treatment with the La-1-like peptide agent may then be selected for use in IVF procedures, such as ICSI procedures. According to an embodiment of the present invention, samples of La-1-like peptide reagents may be incubated with testicular sperm samples. The reagents are used in suitable concentrations (eg, 0.01 micromolar to 1 micromolar). Sperm samples are used at an appropriate concentration (eg, one million per milliliter)...

Embodiment 1

[0158] Venom isolation and La-1-like peptide isolation

[0159] venom origin

[0160] Crude venom from scorpions (Maurus palmatus) was obtained from the Alphabiotoxine Laboratory (Montroeul-au-bois, BELGIUM) or Venomtech (Sandwich, Kent, UK). 10 mg of lyophilized crude venom was dissolved in 500 μL of 100 mM ammonium acetate (pH 6.8) and centrifuged at 10,000 g for 10 min. 250 microliters of the supernatant was used for further purification.

[0161] Purification by Size Exclusion Chromatography (SEC)

[0162] Separation was performed on a Superdex_peptide 10 / 300GL gel filtration column, eluting at 0.5 mL / min in 100 mM ammonium acetate (pH 6.8). Fractions were collected every minute and the absorbance was monitored at 225 nm. Fractions were kept at -20°C until bioactivity screening and mass spectrometry analysis.

Abstract

The invention relates to a La-1-like peptide agent that increases sperm motility. It is non-toxic and it may be used to treat male infertility in humans, and also to increase fertility in animals. Itmay be used in artificial reproductive techniques such as in vitro fertilisation (IVF), including intra-cytoplasmic sperm injection (ICSI). The La-1-like peptide agent may also be used in artificial insemination techniques such as intra-cytoplasmic uterine injection (IUI).

Description

field of invention [0001] The invention relates to the treatment of male or male infertility by improving sperm motility through in vitro or in vivo assisted reproductive technology. The invention also relates to increasing the fertility of male animals in animal production methods including artificial insemination. Background of the invention [0002] Infertility affects one in six couples, with one in two cases being a defect in the male partner. [0003] Male or male infertility presents a highly variable phenotype. One of the most common of these is asthenozoospermia, which is characterized by decreased or absent sperm motility [see Publicover SJ, Barratt CL. Sperm motility: things are moving in the lab! Mol Hum Reprod. 2011 Aug;17(8):453-6]. [0004] Assisted reproductive technology (ART) is the term used to describe methods used to assist couples and, depending on the severity of male infertility, includes: Intrauterine insemination (IUI) for men with mild sperm dys...
