Use of small molecule compounds for the preparation of anti-aging medicaments

ActiveCN121197169BPremature agingEfficacy
This invention relates to the field of anti-aging drug technology, specifically to the application of small molecule compounds in the preparation of anti-aging drugs. These small molecule compounds competitively block the interaction between KDM6A and the vacuolar ATPase V0 domain subunit ATP6V0C by targeting a specific small molecule binding pocket of the histone demethylase KDM6A, thereby inhibiting the demethylation activity of KDM6A on lysine residue 36 of ATP6V0C. These small molecule compounds can effectively downregulate aging-related β-galactosidase activity, improve cell cycle arrest, and reverse the senescent phenotype of nerve cells induced by the environmental pollutant GenX, making them particularly suitable for combating premature aging of nerve cells induced by perfluorinated and polyfluoroalkyl compounds such as GenX. This technical solution can solve the technical problem of the lack of drugs with clearly defined components, a clear mechanism of action, stable efficacy, and the ability to specifically alleviate aging in existing technologies, and has ideal prospects for widespread application.

A method for establishing a premature ovarian failure rat model

ActiveCN118872631BOrganic active ingredientsAnimal husbandryPremature agingGranular leucocyte
The application relates to a method for establishing an early ovarian failure rat model, which comprises the following steps: taking healthy rats, and performing gavage on the rats according to a dosage of 10-20 mg / Kg of a phytostilbene preparation, wherein the dosage is the ratio of phytostilbene to the weight of the rats, and obtaining the early ovarian failure rat model after four days of administration. The early ovarian failure rat model is obtained by performing gavage on the rats after the phytostilbene preparation, the model is simple to establish, and the model establishment time is short. Through ovarian pathological observation, it can be seen that the simulated animal model has obvious ovary tissue damage, follicular granulosa cell destruction and apoptosis, and follicle atresia and other symptoms. The obtained model has good stability and repeatability. The phytostilbene is used to successfully establish the early ovarian failure rat model, lays a good foundation for establishing a stable early ovarian failure model for basic research, and provides a reliable method for the construction of the early ovarian failure model.

Dual Roles of DA1-EOD1 Module in Regulating Organ Size and Postharvest Quality in Lettuce (Lactuca sativa)

Leaf development, especially in leafy greens, is crucial for crop productivity. In various plants, DA1, a pivotal ubiquitin receptor that negatively regulates leaf growth, and EOD1 (ENHANCER1 OF DA1), an E3 ubiquitin ligases that interacts with DA1 through its RING domain, influence organ size. In this disclosure, in lettuce (Lactuca sativa), homologs genes were identified for both DA1 and EOD1, and da1 single mutants, da1 / eod1 double mutants, and DA1 overexpressing lines (DA1ox) were created. The da1 / eod1 double mutant exhibited a striking phenotype with larger organ size, accelerated growth, and increased biomass, resulting in higher yields and rapid growth traits pivotal for Controlled Environment Agriculture (CEA). Additionally, the da1 / eod1 double mutant displayed a pronounced ‘stay-green’ phenotype, characterized by elevated chlorophyll b levels, which implies extended shelf life, and enhanced postharvest quality, both traits indispensable for leafy vegetables in CEA settings. In contrast, the DA1ox line exhibited early senescence and diminished organ size. Based on these findings that spotlight the nuanced roles of the DA1-EOD1 module in modulating plant growth phases and its potential to revolutionize breeding strategies for improved yield and postharvest longevity in CEA-cultivated leafy greens, this disclosure provides guide RNAs for gene editing of DA1 and EOD1, and transformed lettuce having improved organ size and postharvest quality.

Use of PWWP3B in preparation of diagnostic products and therapeutic drugs for testicular premature aging or oligoasthenospermia

ActiveCN119391838BPremature agingHereditary Mutation
The application provides application of PWWP3B in preparation of a diagnostic product and a therapeutic drug for testicular premature failure or oligoasthenospermia, and finds that the occurrence of testicular premature failure or oligoasthenospermia is related to abnormal PWWP3B gene or abnormal expression level of a PWWP3B gene coding protein. Therefore, testicular premature failure or oligoasthenospermia can be definitely diagnosed by detecting the PWWP3B gene sequence or the PWWP3B protein expression level. Meanwhile, it is found that a mutation pathogenic gene related to testicular premature failure or oligoasthenospermia contains 7 mutation sites, and based on the above 7 mutation points, a corresponding AAV carrier is constructed for treatment, and the effectiveness of a gene therapy drug for a specific genetic mutation is confirmed.

A mixed cell preparation and its use in the preparation of a medicament for the treatment of premature ovarian failure

PendingCN122075540ASkeletal/connective tissue cellsArtificial cell constructsPremature agingGranular leucocyte
This invention discloses a mixed cell preparation and its application in the preparation of drugs for treating premature ovarian failure. The mixed cell preparation comprises granulosa cells and mesenchymal stem cells, mixed in a certain ratio (preferably 1:1). Granulosa cells are obtained by inducing differentiation from mesenchymal stem cells and express granulosa cell-specific markers (FOXL2, FSHR, etc.), participating in follicular structure reconstruction; mesenchymal stem cells can improve the ovarian microenvironment, inhibit inflammation, and resist fibrosis, providing a suitable "soil" for the survival of granulosa cells. In vitro experiments are expected to show that mixed culture can significantly increase estradiol secretion levels and cell survival rate. Animal models of premature ovarian failure are expected to show that mixed cell therapy can significantly restore the estrous cycle, improve hormone levels, increase the number of follicles at all stages, and improve pregnancy rates, with effects significantly superior to single-cell therapy. This invention utilizes the synergistic effect of the two cell types to achieve dual repair of the "seed" and the "soil," providing a novel treatment strategy for premature ovarian failure.

PendingCN122272558Arecovery functionavoid instabilityPremature agingOvarian function
This invention belongs to the field of animal model technology, specifically relating to a method for constructing an animal model of aplastic anemia and premature ovarian failure (POF) and its application in mesenchymal stem cell therapy. Busulfan and cyclophosphamide were administered intraperitoneally to the animals six times in the following order: first, busulfan was administered three times consecutively, followed by cyclophosphamide three times consecutively, thus establishing an animal model of the dual pathological state of aplastic anemia and POF. Furthermore, based on this animal model, a mesenchymal stem cell therapy based on disease stages was established, achieving significant recovery of ovarian function and providing a reliable technical foundation for clinical translational research on POF in adolescents.

This invention discloses an antibody for prolonging ovarian function, a culture medium, a method for inhibiting primordial follicle activation, and its application. The antibody is a CD138 antibody. In the technical solution provided by this invention, the CD138 antibody can precisely regulate the transition rhythm of primordial follicles from a quiescent state to a developing state by targeting and inhibiting the primordial follicle activation signaling pathway. This regulatory mechanism can reduce the ineffective activation and loss of follicles in early reproductive years, allowing more primordial follicles to remain dormant, thereby prolonging the physiological lifespan of the follicle pool. This provides biological protection for the even distribution of reproductive cell resources throughout the reproductive cycle in female individuals, effectively avoiding the risk of infertility or premature ovarian failure caused by premature follicle depletion.
