Dual Roles of DA1-EOD1 Module in Regulating Organ Size and Postharvest Quality in Lettuce (Lactuca sativa)

Leaf development, especially in leafy greens, is crucial for crop productivity. In various plants, DA1, a pivotal ubiquitin receptor that negatively regulates leaf growth, and EOD1 (ENHANCER1 OF DA1), an E3 ubiquitin ligases that interacts with DA1 through its RING domain, influence organ size. In this disclosure, in lettuce (Lactuca sativa), homologs genes were identified for both DA1 and EOD1, and da1 single mutants, da1 / eod1 double mutants, and DA1 overexpressing lines (DA1ox) were created. The da1 / eod1 double mutant exhibited a striking phenotype with larger organ size, accelerated growth, and increased biomass, resulting in higher yields and rapid growth traits pivotal for Controlled Environment Agriculture (CEA). Additionally, the da1 / eod1 double mutant displayed a pronounced ‘stay-green’ phenotype, characterized by elevated chlorophyll b levels, which implies extended shelf life, and enhanced postharvest quality, both traits indispensable for leafy vegetables in CEA settings. In contrast, the DA1ox line exhibited early senescence and diminished organ size. Based on these findings that spotlight the nuanced roles of the DA1-EOD1 module in modulating plant growth phases and its potential to revolutionize breeding strategies for improved yield and postharvest longevity in CEA-cultivated leafy greens, this disclosure provides guide RNAs for gene editing of DA1 and EOD1, and transformed lettuce having improved organ size and postharvest quality.

Maize leaf aging control gene ZmPLS1 and application thereof

The invention discloses a corn leaf aging control gene ZmPLS1 and application thereof. According to the invention, firstly, a ZmPLS1 natural mutant zmpls1 with premature senility leaves is identified in breeding improved offspring of a corn excellent inbred line Jing 724, and the photosynthetic pigment content of the ZmPLS1 natural mutant zmpls1 is found to be obviously reduced. In order to further verify the function of the ZmPLS1, the invention also constructs a ZmPLS1 knockout corn strain and a ZmPLS1 interference corn strain, and also discovers that the leaves of the ZmPLS1 knockout corn strain and the ZmPLS1 interference corn strain also have premature aging phenotypes and the content of photosynthetic pigments is significantly reduced. The ZmPLS1 protein participates in regulating and controlling the photosynthetic pigment content and the aging process of the corn leaves for the first time. The invention not only has important significance for clarifying the molecular mechanism of corn leaf senescence, but also has potential application value for delaying the senescence process of corn leaves, increasing the yield and improving the quality.

Application of light wave in medical instrument for treating ovarian aging

The propagation direction of guided light waves in vitro faces the ovary of a tested animal, and verification shows that the activity, proliferation and hormone secretion of KGN cells are remarkably protected, meanwhile, H2O2-induced KGN cell apoptosis is inhibited, the unbalance condition of pro-inflammatory and anti-inflammatory cytokines in an aged mouse is recovered, the mitochondrial function of the ovary of the aged mouse is improved, and the curative effect of the ovary of the aged mouse is improved. The characteristics of reproduction, endocrine and the like of naturally aged ovaries are effectively improved, and the ovarian function of reproductive aging (such as premature ovarian failure) can be recovered.

A molecular marker for detecting a premature senescence gene of a crop plant and use thereof

The application provides a molecular marker for detecting a premature senescence gene of crops and application thereof, and belongs to the technical field of crop breeding. The molecular marker for detecting the premature senescence gene of crops can detect the deletion of four bases AACA in the 11th exon of the coding region of the gene LOC_Os03g31550, so as to form a premature senescence related gene caused by frame shift mutation. Research shows that the rice containing the premature senescence related gene has a stable premature senescence phenotype, the leaf tips of the second leaf and the third leaf are dried and the premature senescence phenotype appears on the 10th day after sowing in a field environment, and the aging lasts to the mature period. The photosynthetic pigment content of mesophyll cells is obviously reduced in the tillering period, the chloroplast structure is changed, the light damage is serious, the photosynthesis is weakened, and the uric acid content is reduced. The application provides a molecular basis for further in-depth research on gene regulation of aging, and provides a reference for cultivating new varieties resistant to premature senescence by using the premature senescence gene.
