Abstract

The invention provides a marker for examining pancreatic ductal adenocarcinoma by using a methylation level of a locus CpG and application of the marker. The cfDNA methylation marker provided by the invention can be used for effectively identifying the pancreatic ductal adenocarcinoma; the PDAC methylation marker protected by the patent is remarkably different in level in cfDNA of normal people and ctDNA of PDAC sufferers and thus can be used for judging whether an examined object suffers from the risk of the PDAC or not. The marker can be used for accurately examining actual levels of PDAC methylation markers corresponding to all panels in a sample provided by an examinee; and by employing reagents, panels, adapters and library building flows provided by a kit, a high-richness and high-diversity DNA methylation library can be prepared from a small amount of cfDNA or ctDNA samples, and signal levels of the PDAC methylation markers provided by the patent are assayed accurately and are used for evaluating whether the examined object suffers from the PDAC or not.
