Embodiment

[0042] Human skin fibroblasts, normal tissue, and hypertrophic scar tissue preparations

[0043] Skin tissues used for the experiment were obtained from 3 patients, and IHC was performed using normal and scar tissues of men and women, and the effect of transformation by siRNA was observed using primary fibroblast cells derived from each tissue.

[0044] After washing the tissue with 70% ethanol, the tissue for IHC was isolated by trimming to remove fat and chopping. After further trimming of the remaining tissue, minced and put into a solution mixed with collagenase, trypsin and EDTA, cells were detached at 37°C and 100 rpm. The isolated cells were cultured in F12 medium supplemented with 10% fetal bovine serum (FBS) and gentamycin.

[0045] Comparison of expression of complement proteins by immunohistochemistry (IHC) experiments on hypertrophic scar tissue

[0046]Tissue obtained from biopsy (Biopsy) was put into O.C.T compound (Cell Poth, KMA-0100-00A) and frozen in dry ic...

Abstract

The present invention relates to a pharmaceutical composition for preventing or treating a hypertrophic scar. The present inventors acknowledged that expression inhibition of thioredoxin 5, PRRC1, S100A11, galectin 1, filamin A, eukaryotic initiation factor-5A, annexin A2 and fatty acid binding proteins 5 can be a new target for relieving and treating a hypertrophic scar. According to the present invention, the treatability of a hypertrophic scar is confirmed by producing thioredoxin 5-, PRRC1-, S100A11-, galectin 1-, filamin A-, eukaryotic initiation factor-5A-, annexin A2- and FABP5-specific siRNAs. Therefore, the knockdown of the proteins or genes coding same induces cell death in a hypertrophic scar and reduces collagen expression and thus can be highly useful for scar treatment.

Description

technical field [0001] The present invention relates to a composition for preventing or treating keloid or hypertrophic scar. Background technique [0002] When the deep layer of the dermis is damaged due to surgery or trauma, the collagen in the dermis that maintains the tension of the skin is overproliferated, and the thinned skin is exposed after the wound heals, leaving traces of skin recovery. scar". This is the result of wound healing, or when the function of properly regulating and inhibiting the wound healing process is impaired, the abnormal dense growth of fibrous tissue can generate hypertrophic scars (Hypertrophic scars) or keloids (Keloid). Hypertrophic scars do not extend beyond the wound and fade over time, but keloids have the difference of growing wider than the damaged area over time, invading into normal skin. For the treatment of hypertrophic scars or keloids, attempts have been made including surgery, radiation therapy, steroid therapy, occlusive dress...
