The present invention relates to a method for mass-producing immunosuppressive T cells using human serum albumin. According to the present invention, when peripheral blood stem cells (PBSCs) obtained from individuals administered with granulocyte colony-stimulating factor (G-CSF) are cultured in a medium containing human serum albumin (HSA) or HSA and mercaptoethanol, immunosuppressive T cells having immunosuppressive effects can be mass-produced. It has been confirmed that the immunosuppressive T cells exhibit excellent therapeutic effects against acute graft-versus-host disease. Therefore, the present invention is expected to be useful in the treatment of immune diseases such as acute graft-versus-host disease.

A method for inducing and activating a conventional dendritic cell subset, the method comprising the following steps: A. seeding human peripheral blood stem cells or umbilical cord blood stem cells in a plate, and performing expansion culture using a basal medium supplemented with cytokine 1; B. performing in vitro induced differentiation culture on the cells after the expansion culture using a basal medium supplemented with cytokine 2; C. sorting the differentiated cells to obtain pure cDC1 cells; and D. performing activation culture on the pure cDC1 cells using a basal medium supplemented with a stimulator to obtain activated cDC1 cells. Disclosed herein is a method for producing mature dendritic cells from peripheral blood mononuclear cells (PBMCs). The method includes the steps of, treating the PBMCs with a cultivating medium supplemented with interleukin 4 (IL-4) and granulocyte-macrophage colony-stimulating factor (GM-CSF) to produce immature dendritic cells; and then treating the immature dendritic cells with the cultivating medium supplemented with IL-4, GM-CSF, tumor necrosis factor alpha (TNF-α), and Prostaglandin E2 (PGE2) to produce the mature dendritic cells. According to embodiments of the present disclosure, the PBMCs used in the present method are isolated from a leukocyte concentrate or a cryopreserved peripheral blood stem cells (PBSCs) stock.

The objective of the present invention is to provide a method for effectively isolating and proliferating stem cells from the peripheral blood. A composition according to the present invention comprises interferon-gamma (IFN-γ) so as to increase the adhesion of stem cells isolated from the peripheral blood, and thus increases acquisition yield of the stem cells. Therefore, the composition and the peripheral blood-derived stem cell isolation and culture method, according to the present invention, are established and used to induce differentiation into progenitor cells, which can be differentiated into somatic cells of various target tissues, and are used for prevention or treatment of diseases.
