Embodiment

[0033] (1) Select the diploid red pomegranate for the experiment, and select the adult fruit trees that have entered the fruiting period. When the top branch flower buds start to grow, select 40 healthy top branch flower buds that start to reproduce and grow, the flower bud length is about 0.2cm, remove the redundant tender leaves and flower buds around it, and wrap the flower buds loosely with absorbent cotton. At 9:00 am and 5:00 pm every day, add a few drops of 0.5% dimethyl sulfoxide (DMSO) to the wrapped flower buds dropwise and add a concentration of 0.1% colchicine solution to soak the flower buds in the colchicine solution . The flower buds are treated by dripping treatment, that is, all the absorbent cotton that wraps the flower buds is dripped and infiltrated. It lasts for 6 days. After the treatment, peel off the cotton and rinse it repeatedly with clean water to prevent the flower buds from being affected by the residual liquid medicine.

Abstract

The invention relates to a cultivation method of polyploid pomegranate. The cultivation method comprises following steps: selecting branched flower buds and branches which grow on the top of a diploid plant, inducing the flower to generate polyploid pollen by dropwise adding chemical materials into the flower buds, carrying out hybridizations between induced male flowers and normal female flowers or between induced female flowers and normal male flowers so as to obtain polyploid hybrid fruits, culturing the hybrid seeds in the hybrid fruits so as to obtain polyploid plants, and culturing the polyploid plants in a tissue cultivation mode so as to obtain polyploid pomegranate plants. The cultivation method overcomes the shortage that a large amount of time is consumed by the conventional chromosome reduplication methods. The cultivation method utilizes the fact that the pollen and ovule cells in the reproductive organs are haploids, namely 1n chromosome, during the pomegranate blossom period, uses chemical materials to process the pollen and ovule cells to induce chromosome doubling, thus largely reduces the time of chromosome doubling process, and has the advantages of available materials, simple operation, and high success rate.

Description

technical field [0001] The invention relates to the field of biotechnology breeding, in particular to a polyploid pomegranate cultivation method. Background technique [0002] According to biological principles and experience, when the chromosomes of plants or animals double, the organisms will become larger, and in plants, the plants, flowers, and fruits will all become larger. For example: pomegranate has 9 pairs of chromosomes, that is, n=9, and normal pomegranate chromosomes are 2n=18. When applied to pomegranate, if its chromosomes can be changed to 4n, 6n or even 8n, it is possible to produce extremely large fruits. If the number of chromosome groups of pomegranate can be changed to an odd number, for example: chromosomes become 3n or 5n, etc., sexual reproduction cannot be carried out, which is the so-called seedless fruit. [0003] Before the present invention, in the existing chromosome doubling methods, 2n plant stem cells were treated separately to become 4n or ...
