The expression vector is obtained by introducing a bovine lactoferricin peptide gene into pYES2 plasmids and utilizing a TPI (triosephosphate isomerase) promoter to substitute a GAL1 (galactokinase 1) promoter of the pYES2 plasmids. The pYES2-TPI-LfcinB recombinant plasmids constructed by the preparation method disclosed by the invention can effectively and stably secrete bovine lactoferricin peptide without using an inducer, so that production cost and production complexity are lowered.
